Isolation and Assay of Rabies Serogroup Viruses in CER Cells

Abstract: Infection of CER cell cultures with field strains of rabies virus, ranging from 0 to 5 mouse brain passages, was detected by immunofluorescence within 2–4 days after infection. A fluorescent focus assay for measuring infectivity of seven rabies serogroup viruses was rapid and reproducible. Rabies field strains and other rabies serogroup viruses also induced cytopathic effect, usually on initial passage. The hemadsorption-negative (HAD) plaque test in BSC-1 cells was successfully applied to laborato… Show more

“…In this experimental model the infected cells were not of neuronal origin (Smith et al, 1977), and CER cells are susceptible to rabies virus infection without prior adaptation by serial passages. The next step would be to evaluate the role of gangliosides in the infection of neurons.…”

“…Titration of infectious virus. The concentration of infectious virus was determined on cultures of chick embryorelated cells as described by Smith et al (1977), with minor modifications. Briefly, the cells were grown in a medium containing NaHCO3 (1.2 g/l) and supplemented with 10% FCS, 2 mM-glutamine and antibiotics.…”
