This study was designed to synthesize and evaluate derivatives of scopolamine (1) as acetylcholine esterase and protease inhibitors. Scopolamine (1) was extracted from the aerial parts of Datura innoxia through bioassay guided fractionation. Five different derivatives of scopolamine (1) were synthesized, and identified through spectroscopic studies. Their acetylcholine esterase (AChE) and trypsin inhibitory potentials were determined through standard protocols and evaluated from the perspective of structure-activity relationship.The synthesized scopolamine derivatives (2-6) showed remarkable AChE inhibitory activity, except for scopoline (6). The results showed higher enzyme inhibition potential of the synthesized compounds (2-5) as compared to scopolamine (1). Maximum inhibition was exhibited by scopolamine N -oxide (89.9 ± 1.2%, IC 50 = 37.4 ± 1.1 μ M)), followed by scopolamine sulfonic acid (70.3 ± 0.8%, IC 50 = 46.9 ± 1.0 μ M) and O -methyl scopolamine (66.1 ± 1.2%, IC 50 = 94.7 ± 0.8 μ M). All derivatives showed moderate activity against trypsin; maximum activity was exhibited by 6 (54.0 ± 1.4%) with IC 50 = 621.2 ± 3.7 μ M.

Correlation between ischemia-modified albumin and lipid levels in patients with acute cerebrovascular disease

Abstract. Acute cerebrovascular disease (ACVD) is associated with high morbidity and mortality rates, however, molecular markers to aid in its early detection are lacking. In this study, we examined the correlation between serum ischemia-modified albumin (IMA), a marker used to identify ischemic events in the heart, and blood lipids in patients with ACVD. Serum IMA levels were determined by enzyme-linked immuno sorbent assay, and total cholesterol (TC), low-density lipoprotein (LDL), high-density lipoprotein cholesterol (HDL) and triglyceride (TG) levels were determined biochemically in 62 patients with cerebral infarction (CI), 40 patients with intracerebral hemorrhage (ICH), 18 patients with subarachnoid hemorrhage (SAH) and 100 healthy individuals (controls). Serum IMA levels were significantly higher in each of the ACVD groups compared to the control group (CI, 80.81±11.97 U/ml; ICH, 80.25±10.91 U/ml; SAH, 74.43±11.39 U/ml and control, 41.08±5.10 U/ml; P<0.05). Additionally, serum TC, LDL and TG levels significantly increased, while serum HDL significantly decreased in the ACVD groups compared to the control group (P<0.05). A positive correlation was found between serum IMA levels and serum TC, LDL and TG levels in the ACVD patients, while serum IMA levels were negatively correlated with serum HDL levels (P<0.05). Thus, serum IMA increased following an ACVD event and was closely correlated with changes in the blood lipid levels. These factors, when combined, may allow the development of molecular markers for an earlier diagnosis of ACVD.

AIM: Increased intracranial pressure following trauma and subsequent possible development of brain death are important factors for morbidity and mortality due to ischemic changes. We aimed to establish the role of ischemic modifi ed albumin (IMA) in the early diagnosis of the process, starting with increased intracranial pressure and ending with brain death. MATERIALS AND METHODS: Eighteen Wistar-Albino rats were divided into three groups; control (CG, n = 6), increased intracranial pressure (ICPG, n = 6), and brain death (BDG, n = 6). Intracranial pressure elevation and brain death were constituted with the infl ation of a balloon of a Fogarty catheter in the epidural space. In all three groups, blood samples were drawn before the procedure, and at minutes 150 and 240 for IMA and malondialdehyde (MDA) analysis. RESULTS: Serum IMA levels at 150 and 240 minutes were higher in ICPG than in CG (p < 0.05). IMA levels were similar in ICPG and BDG. Serum MDA levels at 150 and 240 minutes increased in ICPG and BDG groups compared to CG (p < 0.05). MDA levels were similar in ICP and BD groups. CONCLUSION: IMA should be considered as a biochemical parameter in the process starting from increased intracranial pressure elevation and ending at brain death (Tab. 3, Fig. 5, Ref. 31).
