Is pregabalin effective and safe in total knee arthroplasty? A PRISMA-compliant meta-analysis of randomized-controlled trials

Abstract: Background:Pain management after total knee arthroplasty (TKA) varies and has been investigated for years. Pregabalin as an anticonvulsant agent that selectively affects the nociceptive process has been used for pain relief after operation. This meta-analysis was conducted to examine the evidence of pregabalin in TKA.Methods:Systematic searches of all related literatures were conducted using the following databases: MEDLINE, EMBASE, and Cochrane Central Register of Controlled Trials. Only randomized-controlled… Show more

“…Pregabalin is an analgesic anticonvulsant with anxiolytic effects. It is frequently used in the treatment of chronic neuropathic pain but is increasingly used in the treatment of acute pain [17]. As a part of multimodal analgesia, pregabalin reduces opioid consumption and relieves postoperative pain in patients undergoing knee surgery [11].…”

“…Furthermore, studies suggest that pregabalin can also be used in the treatment of postoperative pain [4] . In a meta-analysis study, Han et al [17] performed a comparison of the drugs used for acute pain treatment and concluded that pregabalin, also known as an adjuvant, increases analgesic outcome.…”

“…A cold pack was used on the surgical site and a single dose of 10 mg intravenous dexamethasone was administered when patients arrived inpatient unit [ 9 , 11 ]. Postoperative oral Loxoprofen sodium (Loxonin; 60 mg three times daily) and Pregabalin (Lyrica; 75 mg twice daily) were administered for pain [ 12 , 13 ]. A standard venous thromboembolism prophylaxis protocol combined mechanical and chemical prophylaxis was adopted for all patients [ 9 ].…”
