Irradiated cultured apoptotic peripheral blood mononuclear cells regenerate infarcted myocardium

Abstract: These data indicate that infusion of IA-PBMC cell suspension in experimental AMI circumvented inflammation, caused preferential homing of regenerative EPC and replaced infarcted myocardium.

“…His group was one of the first showing that paracrine factors released from mesenchymal stem cells are able to promote tissue regeneration. In the following years subsequent work from all over the world supported the hypothesis that rather the paracrine factors than the cells were indeed responsible for the beneficial effects (4)(5)(6)(7)(8).…”

“…Most of the studies report the prevention of the disease, which means that apoptotic cells were infused during the triggering stage of the disease (i.e., at immunization with autoantigens or at the time of transplantation) or before the disease occurred. This is true for Type 1 diabetes in nonobese diabetic mice [47], experimental autoimmune encephalomyelitis [12,62,106], arthritis [36,107,108], fulminant hepatitis [109], contact hypersensitivity [110][111][112], acute and chronic graft rejection [86,[113][114][115], hematopoietic cell engraftment [2,14,85,116,117], acute GvHD disease [14] and reduction of infarction size after acute myocardial infarction [118,119]. Thus, it is difficult to transpose this in the clinical setting where, in general, the disease is already present when a therapeutic solution has to be found.…”

“…Instead of interfering with endogenous post-MI processes, it was noted that apoptotic cells may augment regenerative cues. In a rat MI model, freshly isolated peripheral blood mononuclear cells were forced into apoptosis by cesium irradiation [49 ]. Immediately post-MI, infusion with apoptosis-induced cells, as compared with nonapoptotic cells, improved ejection fraction and attenuated infarct size within 6 weeks.…”
