Iron oxide nanoparticle-induced hematopoietic and immunological response in rats

Abstract: The application and use of iron oxide nanoparticless (IONPs) in the biomedical field are steadily increasing, although it remains uncertain whether IONPs are safe or should be used with caution.

“…It is also used as a food additive in iron-fortified drinks and cereals for human consumption [ 2 ] besides numerous industrial uses in wastewater treatment, gas sensing and semiconductors sorbents lubrications, pigments and coatings [ 3 ]. However, the diverse usage of IONPs induces iron overload and the subsequent initiation of oxidative stress in various organs [ 4 ]. Ingestion, inhalation and dermal penetration are the main routes for the entry of IONPs [ 5 ].…”

“…Phagocytosis of silica and iron oxide particles have been reported to activate immune cells towards pro-inflammatory phenotype characterized by secretion of inflammatory cytokines and generation of reactive oxygen and nitrogen species. 18,20 Activated immune cells are thought to have an increased phagocytic ability. Further, the presence of contaminating sources of lipopolysaccharide (LPS) on particles could result in increased uptake.…”

“…8,1013 Additionally, uptake of particles may alter cytokine secretion, chemotaxis behavior, oxidative burst, and nitric oxide generation in these cells. 1320 However, it remains unclear if the uptake of particles would affect the ability of a phagocytic immune cell to subsequently phagocytose and neutralize a pathogen.…”

“…Even FDA approved some of the IONPs, ferumoxytol and ferumoxides, for use in MRI. Since IONPs are superparamagnetic, they can be used for medical imaging or magnetic drug targeting (MDT) [55,56]. Various studies have been reporting the discrepant results on IONP toxicological effects, some claiming that there is no significant toxicity while others reported severe consequences.…”

“…Various studies have been reporting the discrepant results on IONP toxicological effects, some claiming that there is no significant toxicity while others reported severe consequences. A recent study dealt with in vivo toxicity induced by ultrafine IONPs in rats [56]. The results of 4 weeks of exposure to IONPs showed to be decreasing in bone marrow-mononuclear cell proliferation, with high ROS levels, increased inflammatory response, and DNA changes leading to an apoptotic outcome.…”
