Aerial parts of Barleria prionitis Linn. (Acanthaceae) have shown anti-respiratory syncytial virus, anti-arthritic, anti-inflammatory, hepatoprotective, anti-stress, and immunorestorative activities. The two iridoid glycosides, shanzhiside methyl ester (SME) and barlerin, are the active principles of B. prionitis (BP). For the quality control of BP, rapid methods of quantitation are desirable. Therefore, a simple and reproducible high-performance thin-layer chromatography (HPTLC) method for the simultaneous quantification of SME and barlerin in BP was validated. The marker compounds were isolated, purified, and authenticated by spectral analysis. Precoated silica gel 60 F 254 TLC plates were used as a stationary phase and chloroform-methanol in the proportion of 80:20 (v/v) as the mobile phase. The method was validated in terms of linearity, precision, robustness, accuracy, and limits of detection and quantification. Quantification was performed in the absorbance mode at a wavelength of 240 nm using Deuterium lamp. The linearity was found to be in the range of 200-1000 ng spot -1 for both SME and barlerin, with correlation coefficients of 0.9956 and 0.9932, respectively. The percent recoveries for SME and barlerin were in the range of 99.20 to 99.54%, and 98.93 to 99.19%, respectively. Limits of detection and quantification were found to be 12.95 ng and 21.688 ng, and 17.861 ng and 31.051 ng, respectively, for SME and barlerin. The content (% w/w) of SME and barlerin was found to be 4.91 and 4.69, respectively, in methanol extract of BP. Statistical analysis of the data showed that the proposed method is simple, sensitive, precise, robust and reproducible, specific, and can be employed profitably in place of HPLC.

Ayurvedic sciences helped the early humans to overcome chronic dangerous diseases. There are thousands of varieties of herbs and medicinal plants used to overcome such dreadful diseases. The genus Barleria L. belongs to family Acanthaceae, a medicinally significant group of plants having diversified phytochemicals used for different pharmacological properties. It has been utilized since ancient times for medicinal purposes. It has many plant secondary metabolites such as terpenes, flavonoids, lignins, alkaloids, particularly the iridoid glycosides. The secondary metabolites extracted from Barleria spp. show potential pharmacological activities viz., anti-microbial, anti-inflammatory, anti-oxidant, anti-fertility, anti-arthritic and anti-ulcer activity. In view of these, present review is focused on the phytochemistry, pharmacology and tissue culture studies of some of the important species of the genus Barleria L.

Plant tissue culture techniques are used to maintain or grow plant cells, tissues or organs under sterile conditions on a nutrient culture medium of known composition. Plant tissue culture relies on the fact that many plant cells have the ability to regenerate a whole plant. Single cells, plant cells without cell walls (protoplasts), pieces of leaves, stems or roots can often be used to generate a new plant on culture media given the required nutrients and plant hormones. Cultured plant cells can be offered alternative sources over whole plant cultivation for the production of secondary metabolites. The explants, such as plant leaves, stems, roots, meristems etc. were used for cultivation and establishment of plant cell and tissue cultures. Unfortunately, most often plant cell cultures technologies not lead to an enhancement in secondary metabolites production. Therefore, Strain improvement, methods for the selection of high-producing cell lines, and medium optimizations must be considered to improve the production of secondary metabolites. Several types of cell culture methods are being used to produce bioactive compounds. Cell suspension cultures are preferred for production large amounts of cells for quantitative or qualitative analytical studies. This review refers to certain bioactive secondary metabolites produced in the author's laboratory by different tissue cultures techniques. ÖZETBitki dokusu kültür teknikleri bitki hücrelerini, dokularını veya organlarını bilinen bileşenlerde besleyici kültür ortamlarında steril şartlarda korumak ve beslemek için kullanılırlar. Bitki doku kültürleri, birçok bitki hücresinin tüm bir bitkiyi yeniden oluşturma yeteneğine sahip olması gerçeğine dayanır. Tek başlarına hücreler, hücre duvarı (protoplast) olmayan bitki hücreleri, yaprak parçaları, sap veya kökler gerekli besleyiciler ve bitki hormonları verilen kültür ortamlarında yeni bir bitki üretmek için sıklıkla kullanılabilmektedir. Kültüre edilmiş bitki hücreleri, sekonder metabolitlerin üretimi için tüm bitki yetiştirilmesi süresince alternatif bir kaynak olarak ortaya çıkabilir. Bitki yaprakları, kökler, saplar, filizler vb. gibi eksplantlar, bitki hücre ve doku kültürlerinin yetiştirilmesi ve kurulmasında kullanılmaktadır. Maalesef, çoğu zaman bitki hücre kültürü teknolojileri sekonder metabolit üretiminde bir artışa neden olmamaktadır. Bu yüzden suş geliştirme, yüksek üretilebilir hücre hatlarının seçimi için metotlar ve vasat optimizasyonu gibi yöntemler sekonder metabolit üretiminin geliştirilmesi için dikkate alınmalıdır. Biyoaktif bileşiklerin üretimi için birkaç tipte hücre kültürü kullanılmaktatır. Kantitatif ve kalitatif analitik çalışmalarda büyük miktarda hücre üretimi için hücre süspansiyon kültürleri tercih edilmektedir. Bu derleme, farklı doku kültür teknikleri sayesinde laboratuarlarda üretilen belirli biyoaktif sekonder metabolitlere değinmektedir.
