Validated HPTLC method for simultaneous determination of shanzhiside methyl ester and barlerin inBarleria prionitis

Abstract: Aerial parts of Barleria prionitis Linn. (Acanthaceae) have shown anti-respiratory syncytial virus, anti-arthritic, anti-inflammatory, hepatoprotective, anti-stress, and immunorestorative activities. The two iridoid glycosides, shanzhiside methyl ester (SME) and barlerin, are the active principles of B. prionitis (BP). For the quality control of BP, rapid methods of quantitation are desirable. Therefore, a simple and reproducible high-performance thin-layer chromatography (HPTLC) method for the simultaneous qu… Show more

“…After a bibliographic search, it was found that HPLC is the most common reported method for the analysis of shanzhiside esters [21,22,24]. To our knowledge, three reports are known where analysis of B. prionitis has been done by TLC densitometery using the solvent system chloroform-methanol (8:2, v/v) [28][29][30]. The first report was from our laboratory and this solvent system was developed for the estimation of single iridoid marker barlerin and the method was not validated.…”

“…The most prominent compounds existing in species of Lamium genus are iridoids which usually exist as glycosides. According to the results which have been obtained from in vivo, in vitro, and clinical studies, iridoid-rich plants have anticancer, antioxidant, antibacterial, anti-viral, anti-inflammatory, antiarthritic, immunomodulatory, neuroprotective, and wound healing activities (17).…”

“…Its important active principles are shanzhiside methyl ester (SME), barlerin, and squalene apart from phenolics and avonoids. SME is an iridoid glycoside that has been reported to show various bioactivities, such as anti-in ammatory, anti-depressant, anti-diabetic, antioxidant, and neuroprotective effects (Ghule et al 2012). Some of the possible mechanisms of action of SME include inhibiting the production of pro-in ammatory cytokines, stimulating the expression of β-endorphin in spinal microglia, modulating the activity of monoamine oxidase and serotonin transporter, enhancing the insulin signaling pathway, scavenging free radicals, and protecting against neuronal damage (Ghule et al 2020;Sun et al 2022).…”

“…It has multiple industrial applications in different elds such as pharmaceuticals, cosmetics, food supplements and biofuels (Lozano-Grande et al 2018). Both SME (Ghule et al 2012) and squalene (Nidhi et al 2013) have been quanti ed and isolated in B. prionitis using techniques like HPTLC, ash chromatography and GC-MS and structure of these compounds were elucidated with the aid of extensive NMR spectral studies.…”
