“…In agreement with our previous results, application of 5 µM ionomycin induced fusion of 3.5 ± 3.2% of CD63‐labeled lysosomes ( n  = 197 cells; Figure 5a,d). Considering the strong increase of [Ca 2+ ] i by ionomycin which can reflect rather pathological conditions, we next applied “mild” electrogenic calcium ionophore ferutinin (Abramov & Duchen, 2003; Abramov, Zamaraeva, Hagelgans, Azimov, & Krasilnikov, 2001; Zamaraeva et al, 1997). Ferutinin (30 µM) induced fusion of ∼25.3 ± 3.6% of CD63‐expressing intracellular compartments ( n  = 150 cells; Figure 5d).…”

“…The mechanisms involved in the ferutinin-induced NOS activation cannot be established from our present results, but they could be multifactorial. It has been reported that estrogen and membrane activity of aromatic and aliphatic derivatives of sesquiterpen alcohol carotane, isolated from Ferula genus, modulate hormone production via affecting the mobilization of intracellular calcium (Zamaraeva et al, 1997). Ferutinin may activate phospholipase C (PLC), which hydrolyzes phosphatidilinositol(4,5)biphosphate (PIP 2 ), to generate inositol(1,4,5)-trisphosphate (IP 3 ) and diacylglicerol, leading to the release of intracellular calcium stores and to DAG-dependent activation of protein kinase C (PKC) and activation of Ca 2+ channels with the consequent Ca 2+ influx (Berridge et al, 1983).…”

“…Endogenous non-channel ionophores are among the potential means of mediating and/or regulating mitochondrial ionic flux in vivo. 27,28 Ultimately, testing these hypotheses electrophysiologically will require new methodologies capable of detecting ionic currents at the sub-pA level.…”
