Involvement of SRPK1 in cisplatin resistance related to long non-coding RNA UCA1 in human ovarian cancer cells

Abstract: The therapeutic potential of cisplatin in ovarian cancer treatment is restricted by the occurrence of cellular resistance. We aimed to explore the role of SRPK1 in cisplatin resistance related to the long non-coding RNA UCA1 in ovarian cancer cell.Totally, 24 ovarian cancer tissues and 16 normal tissues were used to assess the expression of UCA1 RNA. UCA1 stable transfected SKOV3 cells were established and the ability of cell migration, invasion and cisplatin resistance was assessed. The expression of SRPK1 an… Show more

“…The multidimensional involvement of HOTAIR in drug resistance may provide a big opportunity to target it for cancer therapy. Similarly, it has been reported that UCA1 upregulation promotes OC cellular viability and induces cellular resistance to cisplatin [103]. In the same study, overexpression of the anti-apoptotic protein serine-arginine protein kinase 1 (SRPK1) was observed; however, cisplatin sensitivity was partially restored following SRPK1 downregulation [103].…”

“…In human cell lines, CRISPR-mediated knockout of lncRNAs such as UCA1, AK023948, and lncRNA 21A has been achieved [102]. Of these lncRNAs, UCA1 promoted tumorigenesis and drug resistance in OC cells [76,103], indicating that it can be targeted by CRISPR. Moreover, inhibiting UCA1 expression by double-guided RNA CRISPR/Cas-9 impaired bladder cancer cell phenotypes in vitro.…”

“…An investigation has shown the elevated expression of UCA1 in ovarian cancer tissues and revealed that the expression of UCA1 RNA in SKOV-3 cells could increase the cell migration, invasion, and cisplatin resistance of these cells. Such function of UCA1 has been shown to be mediated via SRPK1 and apoptosis pathway proteins [58] .…”

“…44 UCA1 might play an important cancer-promoting role in human cancers, such as bladder cancer, breast cancer, ovarian cancer, gastric cancer, lung cancer, tongue squamous cell carcinomas, and CRC. 45,46 Especially in CRC, recent preliminary studies suggested that UCA1 can promote growth of CRC cells and the drug resistance of CRC cells to 5-fluorouracil (5-FU) by decreasing apoptosis, in vitro and in vivo; UCA1 can bundle endogenous miR-204-5p and restrain its activity to increase the protein levels of cyclic adenosine monophosphate responsive element-binding protein 1 (CREB1), negatively related to survival time and clinical outcome in CRC patients. 35 FEZ family zinc finger 1 antisense RNA 1 FEZ family zinc finger 1 antisense RNA 1 (FEZF1-AS1), a recently discovered lncRNA, is located on the counterpart of gene FEZF1.…”
