Involvement of prostaglandin E2 synthesis in the intestinal secretory action of Escherichia coli heat-stable enterotoxin II

Abstract: The prostaglandin response of mouse intestinal epithelial cells after exposure to Escherichia coli heat-stable enterotoxin II was examined. The quantity of prostaglandin E2 produced by the intestinal cells was directly related to the dose of heat-stable enterotoxin II. The change in the amount of prostaglandin E2 over time correlated to that of the volume of fluid released into the intestinal lumen. We then demonstrated that administration of heat-stable enterotoxin II into the intestinal loops of mice induced… Show more

“…The contribution of the system involving PGE 2 has been proposed for diarrhea caused by several pathogens including Vibrio cholerae, Escherichia coli, Salmonella species, and Campylobacter jejuni. The diarrhea caused by these pathogens was suppressed by indomethacin, an inhibitor of PG synthesis (13,19,30). Furthermore, Beubler et al reported that CT increases the level of COX-2 protein and that the PGE 2 produced by the COX-2 mediates the secretion of fluids by CT (2).…”

“…Indeed, treatment of rats with ketanserin, an antagonist of serotonin receptors, results in decreased intestinal secretion by STb implying a role of serotonin in the mode of action of STb . Moreover, Fujii et al (1995) demonstrated serotonin release and fluid accumulation is proportional to the quantity of STb used to treat cells. These results are in accordance with the observations made by .…”

“…Alternative mechanism of STb secretion induction PG synthesis inhibitors significantly reduce the response to STb (Hitotsubashi et al, 1992;Fujii et al, 1995;Dubreuil, 1999). Activation of the ENS by PGE 2 and 5-HT was observed for STb (Sears and Kaper, 1996;Dubreuil, 1997).…”
