Involvement of PAF-Acether in Anaphylactic Bronchoconstriction Induced in Guinea Pigs by Aerosolized Antigen

Abstract: Bronchoconstriction following the aerosolisation of PAF-acether or antigen to guinea pigs induces autodesensitization, but the responses to direct spasmogenic substances are not modified. Bronchoconstriction by PAF-acether is not reduced when it is aerosolized to passively sensitized animals previously desensitized by repeated inhalations of the allergen (ovalbumin). In contrast, when passively sensitized animals are initially desensitized to PAF-acether by repeated inhalations of this agonist, ovalbumin aeros… Show more

“…These changes may, at least in the case of PAF, be due to the action of sulphidopeptide leukotrienes and thromboxanes, which have been shown to be released by PAF from non-mast cell populations in human lung tissue (Schleimer et al, 1986a). Furthermore, it has been demonstrated in the guinea-pig lung that repeated challenges with PAF desensitize against a subsequent allergen challenge but not vice versa (Lagente et al, 1988). This indicates that PAF activates other inflammatory cells that are not mast cells and do not respond to allergen.…”

“…Pretolani et al [35] demonstrated that WEB 2086 antagon ized the pulmonary effects of paf-acether and ana phylactic shock in the guinea pig. In fact, a recent pu blication [36] demonstrated that passively sensitized animals which were made refractory to aerosolized paf-acether, also showed a reduced response to aeros olized ovalbumin. These data suggested that pafacether and antigen may interact in the lung.…”

“…Solutions of FMLP (10, 100 and 1000lgml-P during 2min), aspirin (300mgml-1 during 30min) and indomethacin (l00mgmlP1 during 20min) or the appropriate solvents, i.e., saline for FMLP (the stock solution of FMLP was prepared in DMSO at final concentration of 50 mg ml -1 and then diluted in saline to obtain 1000 ug ml -1) and aspirin, or NaOH (pH 7.8-8.0) for indomethacin (Boukili et al, 1986), were placed in the reservoir of a nebuliser connected to an air compressor, and the flow was directed into the outflow tube of the respiratory pump as described by Lagente et al (1988). FMLP was delivered 5 and 15 min after administration of aspirin and indomethacin intravenously or by aerosol, respectively.…”
