Involvement of Locus Coeruleus Noradrenergic Neurons in Supraspinal Antinociception by α,β-Methylene-ATP in Rats

Abstract: Abstract. We reported previously that intracerebroventricular (i.c.v.) administration of P2X-receptor agonists produced antinociception and the effect was attenuated by i.c.v. pretreatment with b 2 -adrenergic receptor antagonists. The present study examined the involvement of noradrenergic neurons arising from the locus coeruleus (LC) in the supraspinal antinociception by the P2X-receptor agonist a ,b-methylene-ATP in rats. We found that pretreatment with DSP-4 (50 mg / kg, i.p.), which is a neurotoxin to sel… Show more

“…Antinociceptive effects in peripheral tissues were registered after bilateral microinjecton of α,β-meATP into the nucleus locus coeruleus [138] and intracerebroventricular instillation of α,β-meATP [139]. Furthermore, intracerebroventricular administration of the selective P2X 3 receptor antagonist A-317491 or pretreatment with antisense oligonucleotides for the P2X 3 gene decreased inflammatory nociceptive behaviors induced by intraplantar injection of formalin and intraperitoneal injection of acetic acid [139].…”

“…In addition, high levels of 26RFa receptors have also been observed in various brain nuclei involved in the processing of pain such as the parafascicular thalamic nucleus (Weigel and Krauss, 2004), the locus coeruleus (Fukui et al, 2004;Imbe et al, 2004), the dorsal raphe nucleus (Foo and Mason, 2003;Cucchiaro et al, 2005), and the parabrachial nucleus (Cortelli and Pierangeli, 2003;Richard et al, 2005). The NPFF2 receptor has been previously reported also to be present in these nuclei (Elshourbagy et al, 2000;Zeng et al, 2003;Gouarderes et al, 2004), and the role of NPFF, one of the endogenous ligand of NPFF2 (Engström et al, 2003), in the modulation of pain transmission is now widely accepted (for review see Mollereau et al, 2005).…”

“…A recent study determined P2X 1 , P2X 3 , and P2X 2/3 receptors involvement in carrageenan-induced TMJ inflammatory hyperalgesia (18). Conversely, there are data suggesting that P2X purinoceptors, especially P2X 3 and P2X 2/3 , could be involved in antinociception (19,20). …”
