A B S T R A C TVirus infection can result in the alteration of physiological, biochemical and metabolic processes within plants leading to symptom development. Banana bunchy top virus (BBTV) is one of the most destructive viral diseases in Tropical Asia, Pacific Indian Oceania (PIO) regions and Africa leading to 100% yield loss in banana and plantains. Though molecular characterization and their diversity were studied in depth in recent years, information on physiological and biochemical changes during banana-BBTV interaction is still not convincingly explained. Therefore, the present investigation was conducted to find out the quantifiable changes in physiological and biochemical parameters such as proteins, pigment and carbohydrate content, phenolic compounds, polyphenol oxidase (PPO), peroxidase (POX), ascorbate peroxidase (APX), guaiacol peroxidase (GPX), catalase (CAT) and superoxide dismutase (SOD) activities in leaves of banana cultivars Grand Nain (AAA) and Virupakshi (AAB). The amount of carbohydrate contents, phenolic compounds, PPO, POX, APX, GPX, CAT were significantly higher in BBTV infected leaves of both the cultivars over the healthy, whereas total protein content, pigments and SOD activity showed an opposite trend. Overall the results suggest that BBTV infection induces significant changes in enzyme levels leading to irreversible symptom development. Further studies would lead to identification of biochemical markers for studying plant-virus compatible and incompatible interactions.

Plant Growth Promotion and Suppression of Bacterial Leaf Blight in Rice by Inoculated Bacteria

The present study was conducted to evaluate the potential of rice rhizosphere associated antagonistic bacteria for growth promotion and disease suppression of bacterial leaf blight (BLB). A total of 811 rhizospheric bacteria were isolated and screened against 3 prevalent strains of BLB pathogen Xanthomonas oryzae pv. oryzae (Xoo) of which five antagonistic bacteria, i.e., Pseudomonas spp. E227, E233, Rh323, Serratia sp. Rh269 and Bacillus sp. Rh219 showed antagonistic potential (zone of inhibition 1–19 mm). Production of siderophores was found to be the common biocontrol determinant and all the strains solubilized inorganic phosphate (82–116 μg mL-1) and produced indole acetic acid (0.48–1.85 mg L-1) in vitro. All antagonistic bacteria were non-pathogenic to rice, and their co-inoculation significantly improved plant health in terms of reduced diseased leaf area (80%), improved shoot length (31%), root length (41%) and plant dry weight (60%) as compared to infected control plants. Furthermore, under pathogen pressure, bacterial inoculation resulted in increased activity of defense related enzymes including phenylalanine ammonia-lyase and polyphenol oxidase, along with 86% increase in peroxidase and 53% increase in catalase enzyme activities in plants inoculated with Pseudomonas sp. Rh323 as well as co-inoculated plants. Bacterial strains showed good colonization potential in the rice rhizosphere up to 21 days after seed inoculation. Application of bacterial consortia in the field resulted in an increase of 31% in grain yield and 10% in straw yield over non-inoculated plots. Although, yield increase was statistically non-significant but was accomplished with overall saving of 20% chemical fertilizers. The study showed that Pseudomonas sp. Rh323 can be used to develop dual-purpose inoculum which can serve not only to suppress BLB but also to promote plant growth in rice.

Leaf curl disease caused by Cotton Leaf Curl Burewala virus (CLCuBuV) has been recognized as serious threat to cotton in Indian subcontinent. However, information about cotton-CLCuBuV interaction is still limited. In this study, the level of phenolic compounds, total soluble proteins, and malondialdehyde (MDA) and the activities of phenylalanine ammonia-lyase (PAL), peroxidase (POX), catalase (CAT), proteases, superoxide dismutase (SOD), and polyphenol oxidase (PPO) were studied in leaves of two susceptible (CIM-496 & NIAB-111) and two resistant (Ravi and Co Tiep Khac) cotton genotypes. Disease symptoms were mild in the resistant genotypes but were severe in highly susceptible genotypes. The results showed that phenolic compounds, proteins, PAL, POX, CAT, proteases, SOD, PPO, and MDA play an active role in disease resistance against CLCuBuV. The amount of total phenols, proteases, MDA, and PPO was significantly higher in leaves of CLCuBuV-inoculated plants of both resistant genotypes as in non-inoculated plants, and decreased in CLCuBuV-inoculated plants of both susceptible genotypes over their healthy plants. POX, protein content, SOD, and PAL activities showed lower values in resistant genotypes, while they decreased significantly in susceptible genotypes as compared to the noninoculated plants except PAL, which showed non-significant decrease. CAT was found to be increased in both susceptible and resistant genotypes with maximum percent increase in resistant genotype Ravi, as compared to non-inoculated plants. The results showed significantly higher concentrations of total phenols and higher activity of protease, MDA, SOD, and PPO in resistant genotype Ravi after infection with CLCuBuV, suggesting that there is a correlation between constitutive induced levels of these enzymes and plant resistance that could be considered as biochemical markers for studying plant-virus compatible and incompatible interactions.
