Investigation of Microalgal Carotenoid Content Using Coherent Anti‐Stokes Raman Scattering (CARS) Microscopy and Spontaneous Raman Spectroscopy

Abstract: The yield of high-value products, such as pigments that could be extracted from microalgae, is affected by various nutritional and physical factors. Consequently, there is a need for fast visualization techniques that investigate the responses of individual microalgal cells to changing environmental conditions without introducing perturbations. Here, we apply CARS microscopy to map the distribution of pigments in the diatoms Ditylum brightwellii and Stephanopyxis turris and report their relative change in resp… Show more

“…Previous reviews provide a good overview of applications and developments in CARS within a biological context (33)(34)(35). CARS images are constructed from spectral signatures of a particular molecule or macromolecule of interest such as pigments (36,37), fats (38,39), or water (40). The collected spectra can be used directly to study the specific molecular species or indirectly to study cellular-or tissue-level processes.…”

“…The collected spectra can be used directly to study the specific molecular species or indirectly to study cellular-or tissue-level processes. Recent direct studies have investigated, for example, fat storage in Caenorhabditis elegans (41) and pigmentation in microalgae (36). Indirect studies have been important for understanding cancer and tumor proliferation (Figure 2) (37)(38)(39).…”

“…Nowadays, the analysis of carotenoids in algae is performed using high-performance liquid chromatography (HPLC). However, it is important to emphasise that RS can be used for a more detailed analysis of carotenoids in algal cells [17,20,24]. In addition, the development of RS, which will be applied to algae in the future, can be used for real-time research of algal combination in nature.…”

“…The change in carotenoid content in diatoms under different light cycles was analyzed by tuning the CARS emission to 1528 cm −1 and Chl distribution was visualized by TPEF at 850 nm. 11 Recently, a HTS-Raman spectroscopy (HTS-RS) platform was developed to identify more than 100 000 single eukaryotic cells. 12 The current work adapted this platform to detect drug-and light-induced changes in pigment composition of individual living Phaedactylum tricornutum cells.…”
