Investigating the role of vitamin C in tomato through TILLING identification of ascorbate-deficient tomato mutants

Abstract: Besides being a model for fleshy fruits and Solanaceae species, tomato represents one of the main sources of ascorbate (vitamin C) in human diet in many parts of the world. Ascorbate fulfills various roles in plants due to its antioxidant potential and to its connection with other metabolic pathways e.g. cell wall biosynthesis. Among the functional genomic tools recently developed in tomato, EMS (ethyl methanesulfonate) mutant collections provide an opportunity for identifying allelic series of mutations in ta… Show more

“…Additionally, a study of cold and light stress in tomato fruit unveils an interaction between temperature and light to modulate synthesis, recycling and oxidation of ASC in fruit (Massot et al, 2013). Light promoted the accumulation of ASC and GSH in tomato fruit, thus supporting the hypothesis of a stimulation in ASC synthesis by light (Gautier et al, 2009; Massot et al, 2012; Baldet et al, 2013; Smirnoff, 2018).…”

“…In addition, according to Gould (1992), cultivars could be considered a significant source of ascorbic acid with contents higher than 200 mgÁg -1 ; on this basis, five of the eight genotypes analyzed in this work [C H , LR H , O N , P N , and R N ] would be considered a significant source of ascorbic acid because they had values >200 mgÁg -1 . At physiological pH, vitamin C exists as monoanion form called ascorbate, which has proven roles in stress tolerance in tomato and fulfills numerous roles, mainly through its antioxidant potential (Baldet et al, 2013).…”

“…Among these were missense, truncation and splice junction mutations in three different genes involved in ascorbate biosynthesis, which resulted in reduced tolerance to high light intensity in the ascorbate-deficient mutants. These results are described in a companion paper (Baldet et al 2013).…”
