Intrapocket Chemotherapy in Adult Periodontitis Using a New Controlled‐Release Insert Containing Ofloxacin (PT‐01)

Abstract: PT-01 is a newly developed water-soluble controlled-release insert containing ofloxacin (OFLX) as an antibacterial agent. The effect of PT-01 was evaluated by split-mouth application after oral hygiene instruction. No mechanical debridement was carried out during the experiment. PT-01 was applied once a week for 4 weeks in the periodontal pockets of 46 patients with adult periodontitis. The control insert without any antibacterial agent was applied in a selected pocket in a different quadrant for each patient.… Show more

“…gingivalis, and P. intermedia for up to 12 weeks ( with both fast and sustained-release parts containing 10% w/w Ofloxacin, showed that a constant level of above 2 ug/ml, (minimum antibacterial concentration for most pathogenic microorganisms) could be sustained for up to 7 days. Four weekly applications of the insert resulted in significant resolution of periodontal inflammation, PD reduction, and inhibition of supragingival plaque accumulation, compared to control (Table 4) 252 Further investigation failed, however, to show any additional effect on the subgingival microflora beyond SRP alone. 253 Doxcycline.…”

“…Recently Okada and coworkers62 64 have reported the incorporation of Ofloxacin into a new soluble sustained-release delivery system (PT-01) for subgingival therapy. Ofloxacin is a quinolone antibiotic modified to include anaerobic bacteria in its antibacterial spectrum.…”

“…The concepts of locally delivering antimicrobial agents to the periodontal pocket were introduced in late 1980. Sustained or controlled release drug delivery systems have revolutionized the treatment of periodontal disease [175][176][177][178]. Inflammatory pathways considered to be the important criteria in periodontal tissue destruction.…”
