Intranasal Delivery of Mesenchymal Stem Cells Significantly Extends Survival of Irradiated Mice with Experimental Brain Tumors

Abstract: Treatment options of glioblastoma multiforme are limited due to the blood-brain barrier (BBB). In this study, we investigated the utility of intranasal (IN) delivery as a means of transporting stem cell-based antiglioma therapeutics. We hypothesized that mesenchymal stem cells (MSCs) delivered via nasal application could impart therapeutic efficacy when expressing TNF-related apoptosis-inducing ligand (TRAIL) in a model of human glioma. ¹¹¹In-oxine, histology and magnetic resonance imaging (MRI) were utilized … Show more

“…[ 11,35 ] We have shown the homing property of the MNPs labeled MSCs in LPS induced infl ammation model and also in subcutaneous prostate tumor model for both small tumor (≈50 mm 3 ) as well as large tumors (≈500 mm 3 ). This indicates its potential application for diagnosis and treatment of both primary tumors as well as smallsized metastatic tumors.…”

“…Example is in the case of brain tumors in which the intra nasal route of SC is now becoming a novel route of administrations [209,210]. In cancer bearing mice another strategy is introduced to encapsulate the SC in vivo studies.…”

“…Coculture of GBM43 and U87 glioma cells with MSCs-TRAIL displayed notable rise in caspase-3 activity. Survival rate of tumor-bearing mice was enhanced intranasally delivered with MSCs-TRAIL [25]. Carbenoxolone (CBX), a derivative of 18-glycyrrhetinic acid, has been shown to effectively enhance killing activity of TRAIL-express-ing MSCs.…”
