Intracellular macrophage uptake of rifampicin loaded mannosylated dendrimers

Abstract: The present study was aimed at developing and exploring the use of mannosylated dendritic architecture for the selective delivery of an anti-tuberculosis drug, rifampicin (RIF) to alveolar macrophages (AM). The mannosylated dendritic architecture was synthesized and characterized by using IR and NMR spectroscopy. RIF was efficiently loaded into mannosylated dendrimer using dissolution method. Various physicochemical and physiological parameters such as UV, SEM, DSC, drug loading, solubilization, pH dependent i… Show more

“…We attribute the enhanced toxicity attenuation properties of the sialic acid containing polymers to the increased sialic acid valency in these materials. There are many examples of the role of valency in adhesion of ligands to multivalent materials [48][49][50][51][52][53]. More specific to Aβ, others have shown for biological membranes that Aβ binding affinity to sialic acid containing gangliosides increased in clustered or multivalent regions of the membrane compared to membrane regions where sialic acids or gangliosides were unclustered [25,30].…”

“…50% (free RIF) to 85% ( RIF dendrimers). The phagocytic uptake of RIF and RIF-loaded dendrimers in alveolar macrophages harvested from rat lungs showed a clear increase in the intracellular concentration of the antibiotic [101]. A more recent reports investigated 4G and 5G PEGylated-PPI dendrimers for sustain the delivery of RIF.…”

“…In our previous study, we explored concentrations as high as 6 per cent [25]. The RIF aqueous solubility was increased up to 5.4 times, this performance being better than that of cyclodextrins [28] and mannose-grafted poly(propyleneimine) dendrimers [29]. Independently of the copolymer molecular weight, the PEG/PCL molar ratio and its concentration, drug-free micelles display a relatively low physical stability [25,30].…”
