The stem nematode, Ditylenchus dipsaci, is a severe pest in European sugar beet production. In France, Germany, and Switzerland, D. dipsaci damage in sugar beet varies among specific geographic areas. In this study, the reproduction potential of four geographically distinct D. dipsaci populations was determined using sterile carrot disc cultures. In addition, virulence and pathogenicity were investigated in-vivo using sugar beet. No difference was found in the reproduction potential on carrot discs, as well as penetration rate in sugar beet seedlings. The reproduction rate in sugar beet tissue was significantly affected by the D. dipsaci population used. The population from Seeland (CH) showed the highest number of nematodes per plant at 60 dpi (21,071.8 ± 5340.0), compared to the three other populations contained 3588.6 ± 3858.3, 5136.9 ± 4950.8, and 3579.7 ± 5174.2, respectively. Furthermore, the reproduction rate of D. dipsaci was negatively correlated with fresh biomass of sugar beets at 60 dpi. Based on these results, the D. dipsaci population “Seeland” is suitable for breeding programs to detect resistance in sugar beet. After selecting candidate genotypes/varieties, these should be further evaluated for their field resistance in their targeted growing regions.

Ditylenchus dipsaci is an important pathogen of onion and garlic and causes reductions in bulb quality. Between 2012 and 2014, 345 garlic bulb samples were tested for Ditylenchus spp. The average incidence of Ditylenchus spp. was 14.1 %. Twenty-five individuals from 31 Ditylenchus spp. populations were selected for identification by morphology, species-specific PCR, and sequencing of the ITS1-5.8S-ITS2 region. Thirty populations were identified by morphology and speciesspecific PCR as D. dipsaci. Sequencing of the ITS1-5.8S-ITS2 region of the ribosomal DNA found the populations were 99.7 to 100 % similar to each other, and 99.3 to 100 % similar to other D. dipsaci reference isolates from France, and California, USA. One population was identified as Ditylenchus sp. and was 97 % s i m i l a r w i t h i n t h e I T S 1-5. 8 S-I T S 2 r e g i o n t o D. destructor. This information will be useful for the monitoring of future pest outbreaks in garlic in New York State, USA, and the effectiveness of management recommendations.

Garlic plant, which has an important place in the world economy as well as in human nutrition, is grown extensively in Turkey. The stem and bulb nematode, Ditylenchus dipsaci (Kühn) is one of the most important biotic stresses and is under quarantine as it significantly interrupts garlic production wherever it emerges. Thus, being up to date regarding the distribution and population of the stem and bulb nematode, which is locally found in garlic cultivation areas, identification of the races found on the garlic plant at the molecular level and knowing the host spectrum of the races found at the region are of great importance. The objective of this study is to perform species diagnosis using the species-specific molecular markers at ITS-rDNA regions of the stem and bulb nematode isolates collected from the garlic cultivation areas in Turkey in 2016 and 2017. In the study, molecular screening data of the nematodes isolated from the plant and soil samples were analyzed phylogenetically. In this study, upon molecular screening using 9 different species-specific SSR and SCAR primers, it was found that 34 nematode samples of 53 are D. dipsaci.
