Intra-articular injection of collagenase induced experimental osteoarthritis of the lumbar facet joint in rats

Abstract: We aimed to establish an animal model to investigate primary osteoarthritis of the lumbar facet joints after collagenase injection in rats and its effects on chondrocyte apoptosis. We hypothesized that osteoarthritic-like changes would be induced by collagenase injection and that apoptosis of chondrocytes would increase. Collagenase (1, 10, or 50 U) or saline (control) was injected into the lumbar facet joints. The histology and histochemistry of cartilage, synovium, and subchondral bone were examined at 1, 3,… Show more

“…The histological findings of severe cartilage degeneration at 28 days compare favorably to those of Tachihara et al [46], who used injection of Complete Freund's Adjuvant, Kim et al [51]—who used monosodium iodoacetate (MIA) and to those of Yeh et al [57], who injected collagenase into a single lumbar facet joint. …”

“…injection of collagenase is an established model of OA that has been predominantly used to study the mechanisms underlying structural joint damage [15-17]. Histopathological alterations of the knee joint very similar to those observed in human OA have been described for this model, particularly in mice [18,19] and rabbits [20], as well as in the lumbar facet joint of rats [21]. …”

“…34 After that, destruction of the extracellular matrix with subchondral bone exposure, subchondral bone sclerosis, and osteophyte formation are seen. 35 Clinical symptoms of OA include pain, inflammation, and a limited range of motion. 36 Administration of the VT-SR at 500 mg/kg for 28 days significantly ameliorated type II collagenase-induced inflammation and pain levels in the rat knee (Fig.…”

“…The calibration equations and correlation coefficients of resveratrol, hopeaphenol, and (+)-e-viniferin revealed linear relationships between the peak areas and concentrations ( Table 2). The respective retention times and contents of resveratrol, hopeaphenol, and (+)-e-viniferin in VT-SR were 30.0 min (1.1 mg/g), 35.1 min (6.7 mg/g), and 40.9 min (5.1 mg/g).…”
