Intestinal Lymphangiectasis and Lipidosis in Rats Following Subchronic Exposure to Indole-3-Carbinol via Oral Gavage

Abstract: To investigate the toxicity and carcinogenic potential of indole-3-carbinol (I3C), the National Toxicology Program has conducted 13-week subchronic studies in Fisher 344 rats and B6C3F1 mice, and chronic 2-year bioassays in Sprague-Dawley rats and B6C3F1 mice. While the chronic study resultsare not yet available, subchronic study results and short-term special evaluations of interim sacrifices in the 2-year rat bioassay are presented. F344 rats were orally gavaged 300 mg I3C/kg body weight 5 days a week for 13… Show more

“…7 ). However, lacteal dilatation is not commonly observed in rats (Boyle et al 2012 ), and the incidence and severity of lesions were dose dependent. In addition, it is judged to be the effect of BPF on the function of the lymphatic vessels to absorb fat-soluble nutrients, the prolongation of the PT affected by fat-soluble vitamin-related coagulation factors, and the decrease in the blood CHO level.…”

“…neoplasia, fibrosis, granulomatous infections. Lymphangiectasis in the small intestine due to the accumulation of large lipid droplets was described in rats administered indole-3-carbinol, possibly due to an impairment of lipid transport in the lacteals ( Boyle et al 2012 ). Although there were vacuolated macrophages and giant cells in the lamina propria, the epithelial cells were unaffected ( Boyle et al 2012 ).…”

“…A similar histological finding was described in an NTP study of Fischer 344 (F344/N) rats administered indole-3-carbinol via oral gavage. 157 These animals demonstrated a dose-related dilatation of lymphatics (lymphangiectasis) of the duodenum, jejunum, and mesenteric lymph nodes. Material within dilated lacteals stained with Oil Red O and Sudan black, consistent with lipid accumulation.…”
