Interplay between apoptosis and autophagy in colorectal cancer

Abstract: Autophagy and apoptosis are two pivotal mechanisms in mediating cell survival and death. Cross-talk of autophagy and apoptosis has been documented in the tumorigenesis and progression of cancer, while the interplay between the two pathways in colorectal cancer (CRC) has not yet been comprehensively summarized. In this study, we outlined the basis of apoptosis and autophagy machinery firstly, and then reviewed the recent evidence in cellular settings or animal studies regarding the interplay between them in CRC… Show more

“…It is well known that apoptosis is a strictly programmed cell death process, which serves a critical role in maintaining the balance between cell survival and death (18). Normally, apoptosis is a critically regulated physiological process; however, abnormal cellular proliferation and accumulation of genetic defects may occur in instances of impaired apoptotic mechanisms, which could further lead to tumorigenesis and resistance to treatment (19).…”

“…Phase I and II clinical trials have already evaluated the efficacy of the combination of HCQ and chemotherapy ( e.g ., oxaliplatin, fluouropirimidines) and anti-angiogenic agents ( e.g ., bevacizumab) in mCRC patients. Furthermore, mCRC patients have achieved disease stabilization after combining HCQ with vorinostat[ 11 ]. Further elucidation of the effect of the currently existed as well as developing autophagy inhibitors in CRC patients is of paramount importance due to the dual role of autophagy in CRC.…”

“…Growing bodies of evidence have described cell death associated with autophagic features in response to various antitumor agents [ 17 , 18 ]. Thus, the intricate interplay between apoptosis and autophagy is considered to be two different death modalities that serve pivotal roles in the development of a therapeutic strategy for cancer treatment [ 19 ].…”
