The genome size in turtles and crocodiles is thought to be much larger than the 1.2 Gb of the chicken ( Gallus gallus domesticus , GGA), according to the animal genome size database. However, GGA macrochromosomes show extensive homology in the karyotypes of the red eared slider ( Trachemys scripta elegans , TSC) and the Nile crocodile ( Crocodylus niloticus , CNI), and bird and reptile genomes have been highly conserved during evolution. In this study, size and GC content of all chromosomes are measured from the flow karyotypes of GGA, TSC and CNI. Genome sizes estimated from the total chromosome size demonstrate that TSC and CNI are 1.21 Gb and 1.29 Gb, respectively. This refines previous overestimations and reveals similar genome sizes in chicken, turtle and crocodile. Analysis of chromosome GC content in each of these three species shows a higher GC content in smaller chromosomes than in larger chromosomes. This contrasts with mammals and squamates in which GC content does not correlate with chromosome size. These data suggest that a common ancestor of birds, turtles and crocodiles had a small genome size and a chromosomal size-dependent GC bias, distinct from the squamate lineage.

ChickRH6: a chicken whole-genome radiation hybrid panel

-As a first step towards the development of radiation hybrid maps, we have produced a radiation hybrid panel in the chicken by fusing female embryonic diploid fibroblasts irradiated at 6 000 rads with HPRT-deficient hamster Wg3hCl 2 cells. Due to the low retention frequency of the chicken fragments, a high number of clones was produced from which the best ones were selected. Thus, 452 fusion clones were tested for retention frequencies with a panel of 46 markers. Based on these results, 103 clones with a mean marker retention of 23.8% were selected for large scale culture to produce DNA in sufficient quantities for the genotyping of numerous markers. Retention frequency was tested again with the same 46 markers and the 90 best clones, with a final mean retention frequency of 21.9%, were selected for the final panel. This panel will be a valuable resource for fine mapping of markers and genes in the chicken, and will also help in building BAC contigs. chicken / radiation hybrid / mapping

Skeletal muscle satellite cells, a postulated multipotential stem cell population, play an essential role in the postnatal replenishment of skeletal muscles. In the present research, the skeletal muscle satellite cells were isolated from the pectorals of 15-day-old Beijing Fatty Chicken embryos using combined enzymatic digestion of 0.1% collagenase 1 and 0.25% trypsin. Myogenic markers such as MyoD, Pax7 and demin were detected, indicating their skeletal muscle satellite cell identity. Karyotype analysis showed that these in vitro cultured cells were genetically stable. Being exposed to bone morphogen and adipogenic factors, it was proved that they differentiated into osteocytes and adipocytes correspondingly.
