International System for Standardized Avian Karyotypes (ISSAK): standardized banded karyotypes of the domestic fowl (Gallus domesticus)

Abstract: The chicken genetic map is becoming very detailed. The genetic and physical maps need to be integrated in more detail. It is important to have a consensus banded karotype to permit this integration. An international committee met to develop a karyotype for the eight largest chromosomes and the Z and W chromosomes of the domestic fowl (Gallus domesticus). This map is presented in this report.

“…Each peak in the flow karyotype of GGA, TSC and CNI was sorted and the identity of chromosome-specific DNA was verified by hybridization to respective metaphases following the chromosomal idiogram for GGA [15] and karyotypes for TSC and CNI [2]. The identity of GGA chromosomes in the remaining peaks was determined by PCR analysis of chromosome-specific DNA using primer sets [16].…”

“…Other reasons include the importance of the chicken in evolutionary studies as a model organism for birds, and in developmental biology. Also, the chicken has the typical genome organisation of birds, with two chromosome subtypes: nine pairs of cytogenetically distinguishable macrochromosomes including the two sex chromosomes Z and W [19] and 30 pairs of small, cytogenetically indistinguishable microchromosomes. The female is the heterogametic sex (ZW) and the male is the homogametic sex (ZZ).…”

“…The molecular markers for identifying quiescent satellite cells include VCAM-1 (Teboul et al, 1995), c-Met (Ross et al, 2000), Pax7 (Ladjali-Mohammedi et al, 1999), and M-cadherin while the molecular markers Myf5, MyoD and Desmin (Pittenger et al, 1999) can be used to identify proliferating skeletal muscle satellite cells. In this study, we detected the expression of Pax7, c-Met, Demin and MyoD to identify chicken skeletal muscle satellite cells by immunofl uorescence and RT-PCR.…”
