Blueberries are valued for their taste and their high nutritional benefits, including their antioxidant and antiinflammatory properties. In vitro culturing is an alternative method for clonal propagation, and has been used in many biotechnological studies. Most blueberry research is concentrated on highbush and lowbush taxa (Vaccinium corymbosum and Vaccinium angustifolium respectively), with only limited investigations of rabbiteye cultivars (Vaccinium virgatum) that are more suitable for subtropical climates and regions with warmer winters as a result of climate change. There is therefore a need to determine in vitro protocols for that species and group of cultivars. We examined here adventitious shoot regeneration in the 'Delite' rabbiteye blueberry cultivar. Leaf explants were cultured in vitro in Woody Plant Medium (WPM), and the effects of different thidiazuron (TDZ) concentrations, the orientation of the leaf (adaxial or abaxial surface in contact with the medium), and two portions of the leaf segment (basal or apical) were examined. De novo shoot development was studied using light and scanning electron microscopy. All concentrations of TDZ used showed similar survival and regeneration rates; 0.5 μM TDZ showed high efficiency in regenerating adventitious shoots (100%, with 57 adventitious shoots/ explant), as did the adaxial surface in contact with the medium using either the apical or the basal portion of the leaf (97% shoot regeneration, 47.5 adventitious shoots/explant). Anatomical analyses showed direct and indirect organogenesis. The shoots developed leaf primordia with stomata, trichomes, and well-developed vascular tissues, with further elongation and rooting of the plants. We therefore describe here a high-efficiency regeneration method through de novo shoot organogenesis using TDZ in foliar explants of rabbiteye blueberry, with direct and indirect organogenesis.

Southern highbush blueberry plants (Vaccinium corymbosum hybrid) from a commercial blueberry farm were placed in quarantine glasshouses and either exposed to adults of the western golden-haired blowfly Calliphora albifrontalis (Malloch) or no insects at all over a 21-week period. Laboratory reared C. albifrontalis were regularly released into one house to maintain a population of 1000-1500 flies. Flies could only obtain sugar from the plant flowers, and there was sufficient water from the leachate fraction draining out of each bagged plant. Berries were harvested at least twice weekly from the bushes over 21 weeks (46 harvest dates), and yield (both total berry weight and numbers) recorded. Temperature and humidity were very similar between the glasshouses, and plants were irrigated daily with 2.5-4 L/plant as per the producer's recommendations. Adult C. albifrontalis required at least 1.5 flowers/fly to survive. Both more berries (17.14 kg from 9108 berries vs. 10.43 kg from 6379 berries) and larger berries (1.88 vs. 1.63 g/berry) were produced by the bushes with adult C. albifrontalis present. Mature berries are ready to pick ≈10 weeks after flower opening. This study showed that yield between the two treatments began to differ 11 weeks after the flies were first released. Berry yield remained higher in the house with flies (11.29 kg from 6177 berries at 1.83 g/berry) compared with those plants without flies (4.98 kg from 3427 berries at 1.45 g/berry. Berry size was positively correlated with seed numbers. This is the first demonstration under controlled conditions of the ability of an Australian calliphorid blowfly to pollinate and increase yield of commercial blueberry bushes.

La viabilidad denota el potencial que tiene una semilla para germinar, la cual está ligada al éxito o fracaso reproductivo de las poblaciones y de esta manera es una de las primeras variables a evaluar cuando se trabaja con semillas. Nuestro objetivo fue determinar el mejor método para evaluar la viabilidad en 17 especies altoandinas; se comparó la viabilidad mediante dos pruebas: Tetrazolio, para la cual se probaron variaciones de concentración, acondicionamiento y tiempo de exposición; y germinación, que se trabajó bajo condiciones controladas a 20/10 °C, fotoperiodo y termoperiodo de 12 horas. Se utilizó la prueba Kruskal- Wallis para evaluar diferencias. Se encontraron diferencias en el valor de viabilidad entre las pruebas. En 11 especies se recomienda la prueba de Tetrazolio, donde la mejor condición de evaluación fue [1-1,5 %] TZ, 40 °C y 24 horas de exposición. En Puya, Hypericum y Gaultheria la prueba de germinación fue mejor para evaluar la viabilidad. La comparación de las pruebas permitió detectar la presencia de dormancia en algunas especies donde se evidenció una alta viabilidad pero baja germinación. Los resultados permitieron determinar el método más confiable para evaluar la viabilidad en las diferentes especies, aportar información sobre sus requerimientos germinativos y potencial para la propagación sexual, así como detectar la posible presencia de dormancia y la necesidad de profundizar en la evaluación de los métodos de ruptura que permitan aumentar los porcentajes de germinación en algunas de las especies.
