The composition, metabolism, and significance of fatty acids in insect biology are addressed. Fatty acids enter a number of metabolic pathways not directly related to energy storage and production; the unifying theme is that the fatty acids are not only structurally altered in these pathways, but that the alterations carry them from one area of biological significance into another. This theme is developed by offering a perspective on fatty acids in insects and then reviewing three major areas: 1) fatty acid composition, 2) biosynthesis of fatty acids (including polyunsaturated fatty acids and characteristics of certain biosynthetic enzymes), and 3) the biological significance of fatty acids. This last section includes discussions of the biochemistry of waxes, pheromones, and prostaglandins and the roles of fatty acids as components of defensive secretions. Little is known at the biochemical level about the regulation of fatty acid metabolism, and it is suggested that work in this area represents another frontier in insect biochemistry.

A new in vitro technique, utilizing ruptured larvae ofDrosophila melanogaster, was employed to study the incorporation of(3)H-acetate into long-chain fatty acids. Preparative gas-liquid chromatography and scintillation spectroscopy were used to determine the relative activity of each fatty acid from total lipid extracts. Quantitative changes were observed in the distribution of label during the course of the incubation times, which ranged from five minutes to nine hours. All fatty acids which were observed to incorporate acetate in previous in vivo studies also showed incorporation of label under these in vitro conditions. It is concluded that this system may be useful for studying aspects on insect metabolism for short intervals of time.

The total lipids, total neutral lipids, and total phospholipids from Aedes aegypti and aedes albopictus cells cultivated in vitro in a medium containing fetal calf serum were analyzed. The mosquito cells were harvested in the logarithmic and stationary phases of growth. The fatty acid profiles of the lipids showed differences during the aging of the cells but not betweeen species. There was an increase in chain elongation and unsaturation of the fatty acids in the stationary phase when compared with the logarithmic phase of growth. The major components of the fatty acid profiles of the cells were 16:0, 16:1, and 18:1 fatty acids. Few similarities were found between the lipid analysis of the mosquito cells and the growth medium.
