Interleukin (IL)-17/IL-22-Producing T cells Enriched Within the Liver of Patients with Chronic Hepatitis C Viral (HCV) Infection

Abstract: In chronic HCV, T cells producing IL-17/IL-22 may home to the liver; however, circulating levels of IL-17 do not correlate with fibrosis.

“…Correlation analysis was performed using Pearson correlation test. Sample size was determined on the basis of recent studies [7,14,15] supporting the hypothesis that the Th17 cell frequency, the variable in our cohort of patients, has likely a mean of 3.2%¡2.0. We hypothesized that diet can decrease the Th17 cell frequency to 2.0%¡0.15, power590%, significance level 50.05; for this reason to perform the study, at least 24 patients per group are required.…”

“…IL-22 is produced mainly by CD3 + T cells, and to a lesser extent by NKT and NK cells. In Hepatitis C Virus (HCV) infection, IL-22 levels are increased in the liver of patients with viral hepatitis (Dambacher et al 2008;Foster et al 2012). CD4 T cells-producing IL-22 as well as IFN-γ are enriched in the liver compared to blood whereas the percentage of IL-22/IL-17-producing CD4 cells does not differ between liver and blood (Kang et al 2012), suggesting a specific role of these IL-22/IFN-γ double-positive cells.…”

“…Therefore the mediators related to NKT cells such as IL-17 could be involved with hepatitis infections (16,18). Based on our findings the levels of plasma IL-17 in chronic HCV patients were higher than healthy individuals although the difference was not significant (17,(26)(27)(28). Of course this data was in accordant with previous Progression of liver fibrosis by increased liver injury may dominate over the effect of α-GalCer on liver fibrosis, leading to stimulatory effects of a single α-GalCer injection on liver fibrosis induced by NKT cells (16,25).…”
