Interleukin-31 promotes fibrosis and T helper 2 polarization in systemic sclerosis

Abstract: Systemic sclerosis (SSc) is a chronic multisystem disorder characterized by fibrosis and autoimmunity. Interleukin (IL)-31 has been implicated in fibrosis and T helper (Th) 2 immune responses, both of which are characteristics of SSc. The exact role of IL-31 in SSc pathogenesis is unclear. Here we show the overexpression of IL-31 and IL-31 receptor A (IL-31RA) in dermal fibroblasts (DFs) from SSc patients. We elucidate the dual role of IL-31 in SSc, where IL-31 directly promotes collagen production in DFs and … Show more

“… 114 IL-31Ra seems to be upregulated in surrounding tissue cells during the SSc disease state, showing that this axis is likely stimulated in an aberrant manner. 115 IL-33 has been studied as a disease marker in SSc. It follows the disease state of SSc tightly, with increases in affected individuals, and whose drops correlate well with treatment efficacy.…”

“…Overexpression of IL-31 and IL-31 receptors has been shown by Yaseen et al, in the case of SSc. 134 IL-32.-IL-32 is a pro-inflammatory cytokine having a molecular weight of 22 kDa, and its signaling via a mitogen-activated protein kinase (MAPK), p38, and NF-κB are linked with many autoimmune diseases such as inflammatory disorders, RA, IBD, ulcerative colitis, Crohn's disease, etc. 133 It is predicted to have its role against intracellular pathogens by developing resistance against them.…”

“…The present analysis using these new methods has revealed the function of autoantigen-reactive B cells, which had been a black box. These new findings are expected to be applicable to other autoimmune diseases, including PPP, suggesting the possibility of creating a new etiology for autoimmune diseases as a whole, including neutrophils [ 71 ].…”
