Interleukin-1β Activity and Collagen Synthesis in Human Dental Pulp Fibroblasts

Abstract: Immunopathologic reactions play a significant role in inflammatory diseases of dental pulp. Interleukin-1beta (IL-1beta) is recognized as a key player in mediating cellular immune response. In this study, we measured the content of IL-1beta and its effect on collagen synthesis in cultures of fibroblasts derived from healthy and diseased dental pulps. We found that diseased pulp fibroblasts contain 2.5-fold greater amounts of IL-1beta and synthesized 80% greater amounts of collagen compared with healthy pulp fi… Show more

“…In addition, Barkhordar et al (2002) found that diseased pulp fibroblasts contained 2.5-fold greater amounts of IL-1b when compared with healthy pulp fibroblasts. Furthermore, high levels of IL-6 have been connected with the pathogenesis of various inflammatory diseases, such as rheumatoid arthritis (Houssiau et al, 1988) and periodontitis (Reinhardt et al, 1993).…”

“…Thus, the fact that MTA induces neutrophils to produce IL-1β could result in a more organized dental pulp response after the injury, leading to a more favorable pulp repair. 19 All tested materials induced the production of IL-8. This is an important finding, since it is known that all materials can induce different levels of acute inflammation in the pulp.…”

“…18 The production of IL-1β was increased only in the presence of the MTA. In fact, other studies have already reported in vitro 1,[17][18][19][20] and in vivo 12 that MTA is able to improve the production of IL-1β by macrophages, neutrophils and mast cells. This could have implications on the satisfactory effect of MTA after dental pulp exposure.…”

“…Moreover, once activated, fibroblasts may take on new patterns of extracellular matrix remodeling (42,43) or even assume new functions, as in the case of cytokine production (3,44). Smith et al (1) have highlighted differences among fibroblasts from distinct tissues as a possible determinant of local susceptibility to disease.…”
