Interleukin-11 Drives Early Lung Inflammation during Mycobacterium tuberculosis Infection in Genetically Susceptible Mice

Abstract: IL-11 is multifunctional cytokine whose physiological role in the lungs during pulmonary tuberculosis (TB) is poorly understood. Here, using in vivo administration of specific antibodies against IL-11, we demonstrate for the first time that blocking IL-11 diminishes histopathology and neutrophilic infiltration of the lung tissue in TB-infected genetically susceptible mice. Antibody treatment decreased the pulmonary levels of IL-11 and other key inflammatory cytokines not belonging to the Th1 axis, and down-reg… Show more

“…In a mouse model of tuberculosis, IL-11 was increased during infection, and IL-11 inhibition decreased early neutrophil influx and TB disease severity [37, 42]. Here we also demonstrate that neutralization of IL-11 decreases neutrophil accumulation, but this outcome was not associated with changes in bacterial clearance.…”

“…Several studies suggest that systemic IL-11 plays an important role in regulating tissue injury and inflammation during lung infections [10, 11, 17, 37]. Given that many IL-6 family cytokines are induced in murine models of bacterial pneumonia, and modulation of these cytokines results in disparate effects on host defense and resilience to lung injury [2022], we sought to determine functional contributions of IL-11 in response to lung infection.…”

“…In several clinical and experimental studies, IL-11 exhibited anti-inflammatory activity (27,28). IL-11 mediates anti-inflammatory activities during chemotherapy-induced oral mucositis (29), acute intestinal ischemia (30) and in Clostridium difficile toxin A-associated intestinal toxicity (31).…”

“…To test this hypothesis, cytokines and chemokines previously reported to be involved in the host immune response and/or pathogenesis of TB were selected for evaluation. These included cytokines shaping host immunity (IL-1␤, IL-4, IL-6, IL-9, IL-10, IL-12, IL-13, IL-17A, IFN-␥, IL- (such as IL-1␣, IL-6, TNF-␣, IL-8, IL-11, and IL-1␤) (13,15,16).…”
