Interferon-Induced Depression in Chronic Hepatitis C: A Review of Its Prevalence, Risk Factors, Biology, and Treatment Approaches

Abstract: Hepatitis C viral infection is a global health problem that affects approximately 4 million people in the United States. Combination treatment with pegylated interferon (IFN)-alpha plus ribavirin has been shown to be most effective in treating patients with chronic hepatitis C (CHC). Despite its efficacy, one of the most common side effects of this regimen is depression. Whereas IFN-alpha has been found to induce depression in chronic myelogenous leukemia, melanoma, and renal cell carcinoma, CHC patients may b… Show more

“…These neurocognitive dysfunctions have been characterized epidemiologically or pathophysiologically in chronic HCV infection [166,167,176] , and may be explained by the neuroinvasion of HCV, because HCV has been reported to be found in monocytes/microglia of the central nervous system [176,177] . However, antiviral treatment with interferon, particularly interferon-α, is known to exacerbate depression [178] .…”

“…The mechanism of interferon-induced cognitive impairment may be related to prefrontal cortical hypometabolism in addition to damage to hippocampal cells [21]. Ribavirin is a purine nucleoside analogue which may also contribute to interferoninduced depression [22].…”

“…Hepatitis C virus (HCV) infection is a major health problem affecting approximately 170 million people worldwide and 4 million of people in the USA (Asnis & De La Garza, 2006). Up to 85 % of HCV-infected individuals may develop chronic hepatitis C, a disease associated with serious clinical complications, including cirrhosis of the liver and hepatocellular carcinoma.…”
