Pholis fangi is a small, bottom-dwelling fish species endemic to the Yellow Sea and Bohai Sea in China. While previous research has focused on its population biology and molecular structure, little is known about the early developmental stages of this species. In this study, larval and juvenile P. fangi specimens were collected from the Sheyang Sea Area, Jiangsu Province, in 2017. Morphological features were examined using microscopy, and DNA barcoding was conducted to confirm species identification. The research documented detailed changes in yolk sac, fin development, and melanophore distribution patterns across larval and juvenile stages of P. fangi. Comparative analysis with other Pholis species revealed that melanophore distribution is a key distinguishing characteristic, allowing effective differentiation between larval and juvenile stages, as well as between Pholis species. This study provides valuable insights into the early life history of P. fangi, contributing to a better understanding of the genus Pholis. The findings demonstrate the utility of combining traditional morphological observation and molecular techniques for accurate species identification, particularly during the critical larval and juvenile developmental phases.

Fish are among the main target groups when conducting biomonitoring and ecological assessment of aquatic environments. Environmental DNA (eDNA) metabarcoding provides a unique opportunity to enhance biomonitoring throughput, precision, and standardization. However, few fish monitoring studies based on eDNA metabarcoding have been conducted at the basin scale. Taking Dianchi Lake and the three inflowing rivers (Panlong River, Baoxiang River, and Chai River) as research objects, this study monitored fish diversity via eDNA metabarcoding, and the Fish Index of Biological Integrity (F-IBI) was constructed and applied for the ecological assessment of the aquatic environment. Here, we detected 41 species belonging to 9 orders, 15 families, and 35 genera in the sampling areas, including 17 native fish species such as Hemiculter leucisculus, Cyprinus carpio, and Silurus soldatovi. Distinct diversity patterns were observed among Dianchi Lake and the three inflowing rivers. Particularly, the fish diversity of Panlong River was richer than that of Dianchi Lake, Baoxiang River, and Chai River. Water temperature (WT), chemical oxygen demand (COD), total nitrogen (TN), and total phosphorus (TP) significantly influenced the distribution of fish communities in most sampling sites of the Dianchi Lake basin (p < 0.05). Moreover, according to the F-IBI assessment system, 25% of the sampling sites were defined as ‘fine’ or above, and 75% were ‘impaired’ or below. This study provided insights into the ecological restoration and precise management of the aquatic environment of the Dianchi Lake basin.

Establishing a universal indicator of estuary health is an ongoing challenge for coastal ecology that is made more pressing due to the threat that climate change and anthropogenic activity pose to them. Historically, estuarine health was measured through basic physical, chemical, and biological traits, which have been used to routinely monitor estuaries for more than 30 years. However, it is unclear if they are dynamic enough to accurately assess ecosystem health changes driven by the pressures of climate change and anthropogenic activity. Measuring estuarine health indicators and noting incipient indicators relevant to the evolving threats of climate- and anthropogenic-related stressors on estuarine ecosystems is vital for safeguarding them into the future. Monitoring the presence and vitality of disease in estuarine ecosystems may prove to be a significant indicator of estuarine health. Here we review 22 years of literature (354 papers) to identify the role of marine diseases as critical indicators of long-term estuary health compared to traditional methods, with the goal of identifying a key indicator and underlying unifier of other health metrics. As indicators of both general ecosystem health and of multiple other stressors, diseases play a disproportionately significant role in estuary health in the face of climate- and anthropogenic-related stressors. Marine diseases are a unifier of structural and functional estuary health indicators and must be observed and modeled further.
