-Bovine herpesvirus 1 (BoHV-1) has frequently been used as a model for testing parameters affecting DNA immunisation in large animals like cattle. However, the selection of target antigens has been poorly studied, and most of the experiments have been conducted in mice. In the present study, we demonstrated in cattle that a DNA vaccine encoding BoHV-1 glycoprotein gD induces higher neutralising antibody titres than vaccines encoding BoHV-1 gC. Additionally, we show that a DNA vaccine encoding a secreted form of gD induces a higher immune response than a vaccine encoding full-length gD. However, the enhanced immunogenicity associated with the secretion of gD could not be extended to the glycoprotein gC. The current study also describes for the first time the development and the evaluation of a DNA vaccine encoding the major tegument protein VP8. This construct, which is the first BoHV-1 plasmid vaccine candidate that is not directed against a surface glycoprotein, induced a high BoHV-1 specific cellular immunity but no humoral immune response. The calves vaccinated with the constructs encoding full-length and truncated gD showed a non-significant tenfold reduction of virus excretion after challenge. Those calves also excreted virus for significantly (p < 0.05) shorter periods (1.5 days) than the nonvaccinated controls. The other constructs encoding gC and VP8 antigens induced no virological protection as compared to controls. Altogether the DNA vaccines induced weaker immunity and protection than conventional marker vaccines tested previously, confirming the difficulty to develop efficient DNA vaccines in large species.BoHV-1 / DNA immunisation / gC / gD / VP8

CD40-CD154 interactions play an important role in regulating humoral and cell-mediated immune responses. Recently, these interactions have been exploited for the development of therapeutic and preventive treatments. The objective of this study was to test the ability of bovine CD154 to target a plasmid-encoded Ag to CD40-expressing APCs. To achieve this, a plasmid coding for bovine CD154 fused to a truncated secreted form of bovine herpesvirus 1 glycoprotein D (tgD), pSLIAtgD-CD154, was constructed. The chimeric tgD-CD154 was expressed in vitro in COS-7 cells and reacted with both glycoprotein D- and CD154-specific Abs. Both tgD and tgD-CD154 were capable of binding to epithelial cells, whereas only tgD-CD154 bound to B cells. Furthermore, dual-labeling of ovine PBMCs revealed that tgD-CD154 was bound by primarily B cells. The functional integrity of the tgD-CD154 chimera was confirmed by the induction of both IL-4-dependent B cell proliferation and tgD-specific lymphoproliferative responses in vitro. Finally, sheep immunized with pSLIAtgD-CD154 developed a more rapid primary tgD-specific Ab response and a significantly stronger tgD-specific secondary response when compared with animals immunized with pSLIAtgD and control animals. Similarly, virus-neutralizing Ab titers were significantly higher after secondary immunization with pSLIAtgD-CD154. These results demonstrate that using CD154 to target plasmid-expressed Ag can significantly enhance immune responses induced by a DNA vaccine.

ResumenEl siguiente estudio estableció un inmunoensayo enzimático ELISA, usando partículas virales fraccionadas de una cepa de campo del virus IBR, este mostró una alta sensibilidad y especificidad y una baja tasa de falsos negativos y positivos. Los sueros bovinos positivos y negativos, con los cuales se normalizó la técnica de ELISA, fueron analizados por seroneutralización para determinar anticuerpos anti IBR; se recolectaron sueros negativos y positivos con títulos 1:4 a 1:128 y se consideró la dilución del suero bovino 1:50, como la dilución óptima para la técnica de ELISA. Por otra parte, se utilizó el coeficiente de variación BR, para analizar el grado de repetitibilidad de las absorbancias obtenidas de los sueros positivos y negativos, se obtuvo resultados menores del 20%, lo que indico una adecuada repetitibilidad y un bajo coeficiente de variación. La sensibilidad y especificidad encontrada fue de 96 y 98% respectivamente, teniendo como prueba de referencia la seroneutralización. Los resultados obtenidos, determinan que la prueba de ELISA con partículas virales fraccionadas de una cepa de campo, detecta anticuerpos anti IBR con alta sensibilidad y especificidad, siendo una herramienta para el mejoramiento diagnóstico de la enfermedad.Palabras Claves: ELISA, seroneutralización, rinotraqueitis bovina infecciosa, anticuerpos, mejoramiento diagnóstico.
