Interaction of intact type VI collagen with hyaluronan

Abstract: The capacity of non-pepsinyred type VI collagen to oind to hyaluronan wa investigated. Type V! collagen was extracted from bovine meniscal cartilage with 6 M GuHCI and purified by extraction of PEG precipitates and dissociative Sephacryl S-500 HR chromatography. Type VI collagen, detected with a monoclonal antibody, b.Jund in 0.5 M NaCI to hyaluronan-coated micro-wells, the degree of binding being higher at 37°C than 23~C and 4°C. Incubation of type Vl collagen in competitive inhibition assays with testieular … Show more

“…The antibodies used were MAb CS-56 to native CS a, b anti-aggrecan G1 domain c anti-aggrecan G3 domain d, Mab 12C5 to versican G1 domain e and Mab 5D5 to versican G3 domain f aggrecan acting as lubricating agents is a new role for these proteoglycans in the IVD but consistent with roles proposed for CS-PGs in tendon. Furthermore, type VI collagen is capable of interacting with hyaluronan, a well known lubricating component of synovial fluids [28,29,31,44,52,53] and this association may also facilitate the lubrication of collagen bundles.…”

“…[1][2][3][4][5][6] In addition to the skin, hyaluronan is distributed ubiquitously throughout the body, and the list of functions of intra-and extracellular hyaluronan is expanding: vision, cancer, and diŠerentiation, to name a few.…”

“…Intact type VI isolated from bovine cartilage (13), fetal bovine skin (14), and recombinant ␣3 chain A domains (9) were reported to bind hyaluronan. Furthermore, Kielty and co-workers (15) presented evidence for hyaluronan being required as a template for the assembly of type VI collagen tetramers into filaments and for the stability of the filaments once they are formed (15).…”
