Interaction kinetics of peptide lipids-mediated gene delivery

Abstract: Background During the course of gene transfection, the interaction kinetics between liposomes and DNA is speculated to play very important role for blood stability, cellular uptake, DNA release and finally transfection efficiency. Results As cationic peptide liposomes exhibited great gene transfer activities both in vitro and in vivo, two peptide lipids, containing a tri-ornithine head (LOrn3) and a mono-ornithine head (LOrn1), were … Show more

“…To understand the intracellular function of the standardized nanoparticle, ZAPL75C, we have labeled it with FITC-tagged PEG 2000 PE. First, we transfected MDA-MB-468 cells for 24 h with red fluorescent protein (RFP) tagged Rab5a and Rab7a and lamp1 protein using BacMam 2.0 technology to target early endosome, late endosome, and lysosome, respectively. The cells were then cotreated with FITC-tagged PEG 2000 PE-loaded ZAPL75C nanoparticles 30 min prior to fixation. It was seen that both the green and red channels were colocalized for all three cases indicating the effectiveness of the studied nanoparticles to release their payload by all three release pathways.…”

“…Zhao et al . synthesized two peptide lipids containing a tri-ornithine head (LOrn3) and a mono-ornithine head (LOrn1) and studied the interaction kinetics of the liposome-mediated gene delivery ( 68 ). Both lipoplexes were used as vectors to deliver the green fluorescent protein (GFP) into Hela and Hep-2 cells.…”

“…Furthermore, GQDs exhibit high surface area because of their size below 20 nm resulting in higher active sites. According to their unique properties, they have been experimentally and theoretically studied for different applications such as energy storage [3], drug delivery [4], light emitting diodes [5] chemical sensors [6] and biosensors [7]. Nowadays, sensor technology plays an important role within our society, offering a wide range of applications that encompass health monitoring, toxic gas detection, agriculture, and more.…”
