Integrative genome-wide analysis reveals the role of WIP proteins in inhibition of growth and development

Abstract: In cucurbits, CmWIP1 is a master gene controlling sex determination. To bring new insight in the function of CmWIP1, we investigated two Arabidopsis WIP transcription factors, AtWIP1/ TT1 and AtWIP2/NTT. Using an inducible system we showed that WIPs are powerful inhibitor of growth and inducer of cell death. Using ChIP-seq and RNA-seq we revealed that most of the up-regulated genes bound by WIPs display a W-box motif, associated with stress signaling. In contrast, the down-regulated genes contain a GAGA motif,… Show more

“…WIP1-binding sites were mostly identified in promoter and intergenic regions (data S1); a consensus motif (Fig. 3A) computed from these sequences corresponds to the binding site, as previously characterized by Chromatin Immunoprecipitation sequencing (ChIP-seq) and ampDAPseq for three Arabidopsis WIP proteins ( 16 , 17 ). In particular, we found that the CmCRC promoter harbored the consensus CmWIP1 binding motif and was bound by CmWIP1 but not by its zinc-finger variants G242R and S306F (Fig.…”

“…In flowering plants, WIPs are implicated in transmitting tracts, primary root formation, and sex determination ( 22 25 ). Based on their sequence homology and complementation experiments, it has been suggested that WIP genes might show conserved functions across species ( 16 ). Our work reveals the mode of action of WIP proteins.…”

“…Therefore, as the overexpression of all Arabidopsis WIP s has a similar effect on organ and cell size, cell growth modulation might be a common WIP function, at least for the six Arabidopsis WIP s and Gerbera GbWIP2 . The same negative effect on cell growth was also reported recently by [ 13 ] for the Arabidopsis WIP members WIP1 and WIP2 . In global expression analyses of overexpression lines, the authors found gene categories related to stress and cell growth repression and they observed a reduction in plant size in these lines [ 13 ].…”

“…STOP1 (At1g34370) protects plants against acidic soils [ 32 ]. WIP-type AtC2H2-ZFPs, including WIP1 (AT1G34790) and WIP2 (AT3G57670), regulate endothelium differentiation, pollen tube development, and leaf vasculature growth in plants [ 33 , 34 ]. Apart from that, DAZ1 (AT2G17180) and DAZ2 (AT4G35280), which were in clade C-III, are involved in pollen fertility.…”
