Integrated use of LC/MS/MS and LC/Q-TOF/MS targeted metabolomics with automated label-free microscopy for quantification of purine metabolites in cultured mammalian cells

Abstract: Purine metabolites have been implicated as clinically relevant biomarkers of worsening or improving Parkinson's disease (PD) progression. However, the identification of purine molecules as biomarkers in PD has largely been determined using non-targeted metabolomics analysis. The primary goal of this study was to develop an economical targeted metabolomics approach for the routine detection of purine molecules in biological samples. Specifically, this project utilized LC/MS/MS and LC/QTOF/MS to accurately quant… Show more

“…Furthermore, the LC-MS technique has been improved to separate and identify more lipid classes and is currently considered a prominent metabolomics technique for metabolite identification [52]. Faster and more reliable technology for liquid chromatography coupled with quadrupole time of flight mass spectrometry (LC-Q/TOF-MS) has been developed for the simultaneous analysis of a large number of blood metabolites in metabolomics studies [33].…”

“…Purine metabolites are considered clinically relevant biomarkers for improving the progression of PD. 37 Uric acid acts as a powerful antioxidant and an effective free radical scavenger in the human body. Its reduction might promote the occurrence and development of PD.…”

“…In order to address the low fragmentation efficiency of in‐source CID, the high collision energy characteristic of MS ALL technique was applied to LC–Q‐TOF‐MS. LC–Q‐TOF‐MS is a hybrid MS technology consisted of a unit mass resolution quadrupole (Q1), a second quadrupole (Q2), and a TOF mass analyzer with high resolution [211–218]. In the acquisition procedure of MS ALL technique, all the precursor ions are allowed to go through Q1 freely and fragmented in Q2.…”
