Integrated analysis of promoter methylation and expression of telomere related genes in breast cancer

Abstract: Telomeres at the ends of eukaryotic chromosomes play a critical role in tumorgenesis. Using microfluidic PCR and next-generation bisulfite sequencing technology, we investigated the promoter methylation of 29 telomere related genes in paired tumor and normal tissues from 184 breast cancer patients. The expression of significantly differentially methylated genes was quantified using qPCR method. We observed that the average methylation level of the 29 telomere related genes was significant higher in tumor than t… Show more

“…Tel1 promotes MRX recruitment, whereas Rif2 acts as a negative regulator by enhancing ATP hydrolysis that opens Rad50, facilitating MRX release from DNA (184). In line with insights from yeast experiments, changes in expression and posttranslational modification of MRN and telomeric proteins are associated with various diseases in humans (185187). In mouse embryonic fibro-blasts, the MRN complex is required for the response to telomere dysfunction, consistent with the idea that initiating this response shares common elements with the response to interstitial DNA damage (188).…”

“…The on-chip bisulfite conversion was processed in a microfluidic chip which was integrated with the MS-PCR analysis unit, the device has achieved the methylated DNA detection within 80 min with low-input sample (Yoon et al, 2015). These microfluidic chips with capabilities of bisulfite conversion can not only be integrated with self-made testing equipments Heng et al, 2017), but also can be used in combination with commercial testing equipment, for example, the bisulfite conversion of DNA by a microfluidic chip was reported to be subjected to Fluidigm, which successfully accessed multiple CpG loci in multiple DNA samples with high reproducibility (Adamowicz et al, 2018). Researchers also try to use the analysis of DNA melting temperature to determine the methylation of one or more DNA regions.…”
