Insulin Regulates Adipocyte Lipolysis via an Akt-Independent Signaling Pathway

Abstract: After a meal, insulin suppresses lipolysis through the activation of its downstream kinase, Akt, resulting in the inhibition of protein kinase A (PKA), the main positive effector of lipolysis. During insulin resistance, this process is ineffective, leading to a characteristic dyslipidemia and the worsening of impaired insulin action and obesity. Here, we describe a noncanonical Akt-independent, phosphoinositide-3 kinase (PI3K)-dependent pathway that regulates adipocyte lipolysis using restricted subcellular si… Show more

“…Insulin can also stimulate fatty acid synthase (FAS) to produce fatty acids and phosphoenol pyruvate carboxykinase (PEPCK-C) to generate glyceride-glycerol through glyceroneogenesis in adipose, and thus increase triglyceride stores (Holland and Hardie, 1985;Chen et al, 2010). However, fasting decreases the concentrations of both insulin and apelin in circulation (Choi et al, 2010;Yue et al, 2010;Viscarra et al, 2011b;Viscarra et al, 2011a;Yue et al, 2011). Therefore, mammals enduring prolonged food deprivation must possess wellregulated intracellular mechanisms to properly manage lipid metabolism to prevent premature depletion of their energy stores.…”

“…Although Akt activation is both necessary and sufficient to transduce the insulin signal to enhance glucose transport in adipocytes (42), we cannot rule out the involvement of alternate signaling pathways, such as atypical protein kinase C (43). Insulin suppresses lipolysis in WAT via both Akt-dependent and Akt-independent signaling pathways through HSL or perilipin, respectively (44). We did (Fig.…”

“…Karakteristik DM yang lain adalah hambatan lipogenesis dan lipolisis yang berlebihan akibat defisiensi atau resistensi terhadap insulin. 3 Berbagai macam terapi telah ditemukan untuk mengontrol kadar gula darah pada pasien DM. Kontrol gula darah dengan baik akan mencegah kejadian komplikasi penyakit DM.…”
