The effect of elevated CO 2 concentrations on the levels of secondary metabolites was investigated in tobacco plants grown under two nitrogen supply (5 and 8 m M NH 4 NO 3 ) and CO 2 conditions (350 and 1000 p.p.m.) each. High CO 2 resulted in a dramatic increase of phenylpropanoids in the leaves, including the major carbon-rich compound chlorogenic acid (CGA) and the coumarins scopolin and scopoletin at both nitrogen fertilizations. This was accompanied by increased PAL activity in leaves and roots, which was even higher at the lower nitrogen supply. Hardly any change was observed for the structural phenolic polymer lignin and the sesquiterpenoid capsidiol. In contrast, elevated CO 2 led to clearly decreased levels of the main nitrogen-rich constituent nicotine at the lower N-supply (5 m M NH 4 NO 3 ) but not when plants were grown at the higher N-supply (8 m M NH 4 NO 3 ). Inoculation experiments with potato virus Y (PVY) were used to evaluate possible ecological consequences of elevated CO 2 . The titre of viral coat-protein was markedly reduced in leaves under these conditions at both nitrogen levels. Since PR-gene expression and free salicylic acid (SA) levels remained unchanged at elevated CO 2 , we suggest that the accumulation of phenylpropanoids, for example, the major compound CGA and the coumarins scopolin and scopoletin may result in an earlier confinement of the virus at high CO 2 . Based on our results two final conclusions emerge. First, elevated CO 2 leads to a shift in secondary metabolite composition that is dependent on the availability of nitrogen. Second, changes in the pool of secondary metabolites have important consequences for plant-pathogen interactions as shown for PVY as a test organism.

Verticillium wilt is a vascular disease caused by Verticillium dahliae which represents a serious threat for olive growing in Algeria. Many studies have shown the potential involvement of phenolic compounds in the reaction of plants to pathogens. Our study shows that the presence of Verticillium wilt induces a high production of polyphenols in infected olive trees compared to uninfected ones. The presence of high concentrations of flavonoids (3.45%) and alkaloids (0.44%) in the infected trees suggests that flavonoids and alkaloids may play a role in the olive tree resistance to verticillium wilt. The high performance liquid chromatography (HPLC) analysis showed the presence of five phenolic compounds: oleuropeine, luteonine, catechin, and for the first time verbascoside, apigenine-7-glycoside and some derivatives hydroxycinnamic compounds. These substances are good resistance markers and should help to make efficient strategies for the biocontrol of verticillium wilt.

A new phytotoxic protein (cerato-platanin) of about 12.4 kDa has been identified in culture filtrates of the Ascomycete Ceratocystis fimbriata f. sp. platani, the causal agent of canker stain disease. The toxicity of the pure protein was bioassayed by detecting the inducing necrosis in tobacco leaves. The pure protein also elicited host synthesis of fluorescent substances in tobacco and plane (Platanus acerifolia) leaves. We purified the protein from culture medium to homogeneity. Its complete amino acid sequence was determined; this protein consists of 120 amino acid residues, contains 4 cysteines (S-S-bridged), and has a high percentage of hydrophobic residues. The molecular weight calculated from the amino acid sequence agrees with that determined by mass spectrometry, suggesting that no post-transnational modification occurs. Searches performed by the BLAST program in data banks (Swiss-Prot, EBI, and GenBank) revealed that this protein is highly homologous with two proteins produced by other Ascomycete fungi. One, produced during infection of wheat leaves, is codified by the snodprot1 gene of Phaeosphaeria nodorum (the causal agent of glume blotch of wheat), whereas the other is the rAsp f13 allergen from Aspergillus fumigatus. Furthermore, the N terminus of cerato-platanin is homologous with that of cerato-ulmin, a phytotoxic protein belonging to the hydrophobin family and produced by Ophiostoma (Ceratocystis) ulmi, a fungus responsible for Dutch elm disease.The European plane tree (Platanus acerifolia) is an ornamental plant species of the urban environment. A great number of plane trees in the parks and towns of southern Europe have been destroyed by Ceratocystis fimbriata (Ell. and Halst.) Davidson f. sp. platani Walter, the Ascomycete responsible for canker stain disease (1). This disease is characterized by foliar wilting and spreading lesions that involve phloem, cambium, and extensive regions of sapwood (2, 3). The pathogen spreads from tree to tree by means of root grafts of closely spaced plants and, more frequently, through wounds caused by pruning (4).The American species Platanus occidentalis has been shown to contain a source of resistance to C. fimbriata f. sp. platani that could prove of great interest in the genetic improvement of the European plane (5).
