Injection of Human Cord Blood Cells With Hyaluronan Improves Postinfarction Cardiac Repair in Pigs

Abstract: This study using healthy human cord blood mononuclear cells (CB-MNCs) to treat myocardial infarction provides preclinical evidence that combined injection of hyaluronan and human CB-MNCs after myocardial infarction significantly increases cell retention in the peri-infarct area, improves cardiac performance, and prevents cardiac remodeling. Moreover, using healthy cells to replace dysfunctional autologous cells may constitute a better strategy to achieve heart repair and regeneration.

“…In clinical settings, the success of myocardial infarction (MI) treatment depends on ageing, systemic inflammation in host, and processing of cells for infusion. Infusion of human hyaluronan hydrogel coaxed UCSCs in pigs induces angiogenesis, decreases scar area, improves cardiac function at preclinical level, and suggests that the same strategy might be effective for human [ 107 ]. In stem cells therapeutics, UCSCs transplantation can be either autologous or allogenic.…”

“…Animals were sedated with acepromazine-xylazine mix (2 mg/kg). Then, they were anesthetized with zoletil ® (tiletamine and zolazepam commercial mix) at a dose of 4 mg/kg [14,15,16]. A selected dorso-lumbar area was shaved and disinfected with povidone-iodine solution.…”

“…Looking for biomarkers, which can be instrumental in prediction which patients will be responders, the authors suggested some baseline factors, positively associated with both clinical response and retention, such as G-CSF, SDF-1, LIF, MCP-1, and MCP-3. The most recent study described the significant effect of human cord blood mononuclear cells (CB-MNCs) injection for cardiac repair in ischemic heart disease, mainly by promotion of angiogenesis in the infarcted region [49].…”
