In the present study, olive leaf extract, harvested from four seasons, extracted with solvent (methanol and water), was treated by D101 and Sephadex LH-20, and divided into 10 fractions by TLC detection. The antioxidant activity was evaluated using DPPH, superoxide anion scavenging and reducing power assays. Tyrosinase inhibitory activity was also performed with L-DOPA as substrate. On extraction method, there is no significant difference in methanol and water. Among 12 months, autumn showed the best harvest time in whole. OLE showed 90% tyrosinase inhibitory activity and inhibition type was non-competitive competition. By chromagraphy treatment, the ingredients of polysacchmaride and protein showed high bioactivity towards antioxidant and anti-tyrosinase activity, the rest part showed different extent of activity and prove some effect by chromatography treatment indirectly. OLE might be a valuable bioactive source and can be applicable in both the cosmetic and medical use.

Lavandula genus is an important member of Labiatae (Lamiaceae) family. People use commonly Lavandula stoechas as a medicinal plant for various diseases around the world and also in Turkey. The aim of this study was to investigate cytotoxic and genotoxic effects of aqueous extracts (40, 80 and 120 g/L) from L. stoechas flowers on Allium cepa root tip meristem cells. For this purpose, A. cepa onion bulbs were treated with the above-mentioned L. stoechas flower extracts for 72 h. Spring water (pH 7.3) was used as a control. The result of this study sowed that aqueous extracts reduced mitotic index, but induced chromosome aberrations and mitotic aberrations in comparison with control, significantly (p < 0.05). Aqueous extracts induced breaks, stickiness, pole deviations and micronuclei. Furthermore, these effects were related to extract concentrations. These results showed that L. stoechas aqueous extracts have cytotoxic and genotoxic effects.

Tyrosinase inhibitory activity of supernatant and semi-purified extracts from squid pen fermented with Burkholderia cepacia TKU025

Reports of tyrosinase inhibitors from microorganisms are rare. The tyrosinase inhibitory activity was found from the culture supernatant of Burkholderia cepacia TKU025 with 1 % squid pen as the sole carbon/nitrogen source. The fourth-day supernatant showed the strongest tyrosinase inhibitory activity (500 U/ml) and the highest total phenolic content (789 lg/ml GAE). The supernatant was further purified by liquid-liquid partition, and it was found that the methanol extract exhibited the lowest IC 50 value of 2 lg/ml in the tyrosinase inhibitory activity assay and was compared with the positive control kojic acid and vitamin C. Significant associations between the tyrosinase inhibitory potency and the total phenolic content were found for the supernatant, extract, and fractions. The tyrosinase inhibitory activity persisted even when it was treated with acidic or alkaline conditions of pH 2 or 12. The tyrosinase inhibitory activity remained at 100 %, even after treatment at 100°C for 60 min. The tyrosinase inhibitor also showed antimicrobial activity against Escherichia coli, Bacillus cereus, Aspergillus fumigatus, and Fusarium oxysporum.
