Inhibitory effects of Gleditsia sinensis fruit extract on telomerase activity and oncogenic expression in human esophageal squamous cell carcinoma

Abstract: Previous studies have shown that the anomalous fruit extract of Gleditsia sinensis (GSE) exhibited apoptotic properties in various solid and non-solid tumors in vitro. However, the inhibitory actions of GSE on oncogenic expression and telomerase activity in esophageal squamous cell carcinoma (ESCC) have not been studied before. In the present study, the anti-cancer effects of GSE were demonstrated in three ESCC cell lines (HKESC-1, HKESC-2 and SLMT-1) by MTS and anchorage-independent clongenicity assays, expre… Show more

“…We have first explored the oncogenic properties and altered expression of FAM134B (JK1) gene in oesophageal squamous cell carcinoma (Tang et al, 2007). Also, FAM134B has been reported to have roles in pathogenesis as well as prediction of prognosis of other gastrointestinal cancer, namely colorectal cancer (Kasem et al, 2014).…”

“…However, several studies investigated the effect of other herbal extracts on telomerase expression and concentration in PCa cell lines. Fruit extract of Gleditsia sinensis exhibited anti-cancer and telomerase inhibition effects on ESCC (esophageal squamous cell carcinoma) cell line (39). In human lung carcinoma cell line A549, aqueous extract of the Platycodon grandiflorum root represents the apoptotic events with the reduced telomerase activity and downregulation of Bcl2 expression (40).…”

“…Its crude extract showed anti-allergenic (13) and anti-inflammatory effects (14). However, scientific evidence remains unclear for support of the anti-tumor effect of the extract of Gleditsia sinensis thorns, although the fruit extract of Gleditsia sinensis has been credited with tumor growth inhibition in several cell lines (15,16). The present study is the first report of the anti-tumor effect of Gleditsia sinensis thorns on a relevant pathway between the cell cycle regulation and signaling pathways involved in growth inhibition and MMP regulation in human colon cancer HCT116 cells in vitro.…”
