Inhibitory Effect of Tetrandrine on Lens Proteins-Induced Ocular Inflammation in Rabbits

Abstract: Ocular inflammation was induced by 25 ul of lens proteins (62. 5mg/ml) injected into anterior chamber of the rabbit eye. Tetrandrine (Tet) (50mg/kg ip)and Indomethacin (Ind) (20 mg/kg ip) showed marked inhibition on this ocular inflammation. Maximum inhibition rate of Tet and Ind was 65% and 66% and their antiinflammation action lasted 5 and 4 h, respectively. In the early phase of ocular inflammation (at 2 h) the total content of prostagladin E(PGES) in the iris was reduced by Tet and Ind. Ind showed a greate… Show more

“…The lens cells in the anterior chamber also worsen the development of the anterior segment and can delay eye development in general. Moreover, the crystallins in the anterior chamber might induce ocular inflammation and glaucoma ( 50 , 51 ). During this embryonic stage (E15.5) and the early postnatal period (P10) before eye opening, genes representing inflammation markers such as IL-1β and TNF-α are significantly up-regulated suggesting that congenital ocular inflammation occurs.…”

“…Thus, inhibition of IL-1 release and IL-1 receptor should be able to result in an effective and potent suppression of inflammation without producing the serious side effects produced by steroids, NSAIAs, and PAF antagonists (9). It was reported that prostaglandin and leukotriene generation were inhibited by Tet (4,14) which suppressed the release of two major metabolites of arachidonic acid, namely the leukotriene B4 (LTB4) and the thromboxane B2 (TXB2). This was catalyzed by calmodulin-dependent phospholipase A2 (PLA2) from the membrane phospholipids (15).…”

“…In previous publications (4,5), we have reported that Tet is able to inhibit uveitis induced by lens-proteins and bovine serum albumin without producing ocular hypertension and withdrawal rebound which are frequently induced by steroids in rabbits.…”

“…As an anti-inflammatory agent, tetrandrine was shown to block the ocular inflammation induced by lens proteins, bovine serum albumin, endotoxin, and IL-1 (30,31). It was reported that tetrandrine inhibited the generation of leukotriene B4 and thromboxane B2, a prostaglandin, in the arachidonate cascade (31,32).…”

It was reported that tetrandrine inhibited the generation of leukotriene B4 and thromboxane B2, a prostaglandin, in the arachidonate cascade (31,32). Tetrandrine was also able to suppress microvascular leakage and other inflammatory processes induced by PAF and other allergic mediators (33) and to inhibit the capacity of human mononuclear leukocyte's and monocyte's production of TNFa and lymphocyte's release of TNF/3 (34,35).…”
