Inhibitory Effect of Delphinidin from Solanum melongena on Human Fibrosarcoma HT-1080 Invasiveness in vitro

Nowadays, much attention has been paid to the development of anti-inflammatory agents from marine natural resources. As a result of screening anti-inflammatory agents from marine algae using immunoassay, we found for the first time that ethanolic extract of Ishige okamurae (IO) classified into brown algae was effective in inhibiting the production of inflammatory mediators, such as tumor necrosis factor-alpha, interleukin-1beta, interleukin-6 and prostaglandin E(2), in RAW264.7 cells stimulated by lipopolysaccharide, compared with dexamethasone and aspirin used as positive control in this study. Moreover, transcriptional activation of NF-kappaB transcription factor that regulates the expression of these inflammatory mediators was also examined using reporter gene assay and western blot analysis. It was observed that IO extract exerted anti-inflammatory effect via inactivation of NF-kappaB transcription factor in macrophages. In addition, the expression and activity of matrix metalloproteinase-2 and 9 that play an important role in chronic inflammation were decreased in dose-dependent manner in the presence of IO extract in HT1080 cells. The above results suggest that IO extract can inhibit inflammation through inactivation of NF-kappaB transcription factor in macrophage.

Anthocyanidins inhibit migration of glioblastoma cells: Structure‐activity relationship and involvement of the plasminolytic system

Complete resection of malignant glioblastomas is usually impossible because of diffuse and widespread invasion of tumor cells, and complementary approaches need to be developed in order to improve the efficacy of current treatments. Consumption of fruits and berries has been associated with decreased risk of developing cancer and there is great interest in the use of molecules from dietary origin to improve anticancer therapies. In this work, we report that the aglycons of the most abundant anthocyanins in fruits, cyanidin (Cy), delphinidin (Dp), and petunidin (Pt), act as potent inhibitors of glioblastoma cell migration. Dp clearly exhibited the highest inhibitory potency, this effect being related to the ortho-dihydroxyphenyl structure on the B-ring and the presence of a free hydroxyl group at position 3. Dp decreases the expression of both urokinase-type plasminogen activator receptor (uPAR) and the low-density lipoprotein receptor-related protein (LRP), acting at the transcriptional levels. In addition, Dp upregulated urokinase-type plasminogen activator (uPA) and downregulated the plasminogen activator inhibitor-1 (PAI-1) but decreased, in a concentration-dependent manner, the uPA-dependent conversion of plasminogen to plasmin, indicating that the upregulation of uPA observed with these compounds was not associated with induction of the plasminolytic activity. Overall, these results demonstrate that Dp, Pt, and Cy affect plasminogen activation, thus leading to the inhibition of glioblastoma cell migration and therefore they may be helpful for the development of new strategies for cancer prevention and therapy.

Nutritional Quality and Health Benefits of Vegetables: A Review

Vegetables are considered essential for well-balanced diets since they supply vitamins, minerals, dietary fiber, and phytochemicals. Each vegetable group contains a unique combination and amount of these phytonutriceuticals, which distinguishes them from other groups and vegetables whithin their own group. In the daily diet vegetables have been strongly associated with improvement of gastrointestinal health, good vision, and reduced risk of heart disease, stroke, chronic diseases such as diabetes, and some forms of cancer. Some phytochemicals of vegetables are strong antioxidants and are thought to reduce the risk of chronic disease by protecting against free radical damage, by modifying metabolic activation and detoxification of carcinogens, or even by influencing processes that alter the course of tumor cells. All the vegetables may offer protection to humans against chronic diseases. Nutrition is both a quantity and a quality issue, and vegetables in all their many forms ensure an adequate intake of most vitamins and nutrients, dietary fibers, and phytochemicals which can bring a much-needed measure of balance back to diets contributing to solve many of these nutrition problems. The promotion of healthy vegetable products has coincided with a surging consumer interested in the healthy functionality of food. Because each vegetable contains a unique combination of phytonutriceuticals, a great diversity of vegetables should be eaten to ensure that individual's diet includes a combination of phytonutriceuticals and to get all the health benefits. This article makes a review and discusses the nutritional quality and health benefits of the major groups of vegetables. More interdisciplinary work is required that involves nutritional and food scientists as well as others from biomedical fields to ascertain the thrue function of specific phytonutriceuticals.
