Inhibition of Vascular Permeability Increase in Mice

Abstract: Effects of glucocorticoids on IgE antibody-mediated 48-hour homologous passive cutaneous anaphylaxis (PCA) and skin reactions caused by mediator releasers and vascular permeability increasing factors were investigated comparatively. Both PCA and skin reactions were evoked in the mouse ear and these reactions were quantitatively evaluated by measuring the amount of dye extravasated into the ear. The glucocorticoids hydrocortisone, prednisolone and dexamethasone inhibited the PCA significantly. The maximum inhib… Show more

“…Preparation for Anti-OVA Serum The procedure employed was a slight modification of the method of Inagaki et al 15) Ovalbumin was suspended in 20 mg/ml Al(OH) 3 solution (final concentration of OVA, 10 mg/ml), and 0.2 ml of this suspension (OVA, 2 mg) was intraperitoneally injected into 4-week-old female Balb/c mice (Japan SLC, Hamamatsu, Japan) 5 times at 2 week intervals. Ten days after the 5th injection of OVA, whole blood was collected to obtain anti-OVA serum.…”

“…The vascular effect of prednisolone may also participate in its inhibition of swelling. 17) The epidermal permeability barrier is made up of highly hydrophobic lipids, which form multiple membrane bilayers in the outer cornified cell layers. [2][3][4] Patients with atopic dermatitis often exhibit dry skin that may correlate with a defect in the epidermal barrier structure.…”

“…Corticosteroids are potent inhibitors of inflammatory oedema and this probably represents a major component of their therapeutic activity in inflammatory diseases. Corticosteroids, indeed, inhibit skin reactions, caused by vascular permeability increasing factors, including IgE antibody-mediated homologous passive cutaneous anaphylaxis (Inagaki et al, 1988). In rat tracheobronchial mucosa, corticosteroids failed to reduce local oedema induced by cigarette smoke and by instillation of hydrochloric acid and gastric juice Martling & Lundberg, 1988).…”
