Abstract.[Purpose] The purpose of the present study was to investigate the effect of a magnetic infrared laser (MIL) on the 2, 4-dinitrofluorobenzene (DNFB)-induced contact dermatitis, type I allergic mouse model. [Subjects] Male ICR mice.[Methods] Contact dermatitis was induced by sensitization with dinitrophenyl-derivatized ovalbumin (DNP-OVA) and DNFB challenge as antigen. MIL radiation doses of 1.33, 2.66 and 6.65 J/cm 2 were given, once a day for 7 continuous days, to DNP-OVA sensitized mice and compared with 15 mg/kg dexamethasone (intraperitoneal treatment). The edematous changes and scratching behavior were measured together with ear histopathological observations. [Results] Immediately after DNFB challenge to the ear or paw of DNP-OVA sensitized mice, increases of ear and paw thicknesses and weights were detected as well as anterior ear skin (dermis to epidermis) thickness and paw scratching behavior increases. However, these DNFB-induced increases of edematous changes and scratching behavior were significantly and dose-dependently decreased by MIL irradiation.[Conclusion] The results obtained in this study suggest that MIL radiation has relatively favorable effects on allergic dermatitis, and MIL therapy may represent an alternative approach for the treatment of allergic cutaneous diseases.

−The effect of Picrorrhiza Rhizoma (PR) aqueous extracts were evaluated on 2,4-dinitrofluorobenzene (DNFB)-induced contact dermatitis, type I allergic model. Contact dermatitis was induced by sensitization with dinitrophenyl-derivatized ovalbumin (DNP-OVA) and DNFB challenge as antigen. Three different concentrations of PR extracts (300, 150 and 75mg/kg) were orally administered to DNP-OVA sensitization mice once a day for 7 days with reference materials; dexamethasone (15mg/kg, intraperitoneal treatment). End of 7 days oral administration of PR extracts or intraperitoneal treatment of dexamethasone, the changes on the edematous changes and scratching behavior were measured. Immediate after DNFB challenge on ear or paw of DNP-OVA sensitized mice, increases of ear and paw thicknesses and weights were detected with anterior ear skin (dermis to epidermis) thickness and paw scratching behavior increases. However, these DNFB-induced increases on ear and paw thicknesses, weights and scratching behaviors were decreased by treatment of all three different dosages of PR extracts and dexamethasone, respectively. In addition, the increases of anterior skin thicknesses were also dramatically inhibited by treatment of all three different dosages of PR extracts and dexamethasone at histopathological observations. The results obtained in this study suggest that oral treatment of PR extracts also has relatively favorable effects on allergic dermatitis.

Repeated topical application of glucocorticoids augments irritant chemical-triggered scratching in mice

Topical glucocorticoids, widely used for the treatment of a variety of dermatitises, are known to exacerbate atopic dermatitis after long-term or inappropriate use. In some animal models, topical glucocorticoids augment the allergic cutaneous inflammation after repeated application, suggesting a relationship between these and clinical observations. We investigated whether topical glucocorticoids augment itching, rather than inflammation, resulting in the exacerbation of atopic dermatitis. Mice receiving repeated topical application of glucocorticoids, betamethasone valerate or dexamethasone, to the ear for 1 week showed significantly higher scratching frequency after application of an irritant chemical, 2,4-dinitrofluorobenzene (DNFB) or 12-O-tetradecanoilphorbol 13-acetate (TPA) than those receiving either a glucocorticoid or irritant chemical alone. In contrast, the increase in ear thickness induced by application of TPA was significantly suppressed by dexamethasone. Substance P (SP) and nerve growth factor (NGF) levels were higher in the ear receiving betamethasone valerate followed by DNFB application than in that receiving DNFB alone. In addition, histopathological studies revealed an increased density of nerve fibers in the ear receiving betamethasone valerate or dexamethasone followed by DNFB application. Oral administration of betamethasone valerate was not associated with an increase in either scratching frequency or SP or NGF level in the ear. These results suggest that repeated topical application of glucocorticoids may augment irritant chemical-triggered scratching through an increase in SP and NGF levels and nerve fiber density at the application site. These findings might explain the etiology of the exacerbation of atopic dermatitis and other dermatitises, occurring after long-term or inappropriate use of topical glucocorticoids.
