Inhibition of myosin light chain phosphorylation decreases rat mesenteric lymphatic contractile activity

Abstract: Based on the well-established finding that the phosphorylation of myosin light chain 20 (MLC20) plays an essential role in blood vessel smooth muscle contraction, we investigated if phosphorylated MLC20 (pMLC20) would modulate the tonic and/or phasic contractions of lymphatic muscle. The effects of ML-7, a MLC kinase inhibitor (1-10 M), were tested on the contractile parameters of isolated and cannulated rat mesenteric lymphatics during their responses to the known modulators, pressure (1-5 cmH2O) and substanc… Show more

“…The rats were anesthetized with Innovar-Vet (0.3 ml/kg im), which is a combination of a droperidolfentanyl solution (droperidol at 20 mg/ml and fentanyl at 0.4 mg/ml) and diazepam (2.5 mg/kg im). For isolation of a rat mesenteric lymphatic vessel, a 6-to 7-cm-long loop of small intestine was exteriorized through a midline laparotomy as described elsewhere (66). A section of the mesentery was gently positioned over a semicircular viewing pedestal on a vessel preparation board.…”

“…Lymphatic vessel diameter was digitally recorded using established video microscopy techniques to monitor contractile function. 5,[9][10][11]13 Indocyanine Green was purchased from Akorn Inc. (Lake Forest, IL) (catalog # NDC-17478-701-25) as part of the ICG kit (catalog # NDC-17478-70102). ICG was perfused through isolated lymphatic vessels at a transmural pressure of 5 cm H2O and an imposed flow of 5 cm H2O (a pressure/imposed flow profile for which we have performed numerous control and pharmacological treatment studies.…”

“…Subsequently, we calculated these indices for the assessment of isolated lymphatic contractility as described previously [7][8][9] , including the TI, which refers to the preparing status of lymphatic subsequent contraction, CA, which refers to the normalized intensity of lymphatic contraction, FPF, which refers to the capacity of lymph transport. We found that hemorrhagic shock lead a decrease in CF, TI, and FPF of isolated lymphatics from thoracic duct, suggesting a reduction of lymphatic contractility.…”
