Inhibition of gastric emptying and intestinal transit by amphetamine through a mechanism involving an increased secretion of CCK in male rats

Abstract: 1 The e ect of amphetamine on gastrointestinal (GI) transit and the plasma levels of cholecystokinin (CCK) were studied in male rats. 2 Gastric emptying was inhibited both acutely and chronically by the administration of amphetamine. GI transit was decreased by the acute administration of amphetamine but not a ected by the chronic administration of amphetamine. 3 Plasma CCK levels were increased dose-dependently by amphetamine. 4 Proglumide, a CCK receptor antagonist, prevented amphetamine-induced inhibition o… Show more

“…Gastric emptying was measured as described by Doong et al [8]. Rats were intubated via a catheter (PE-205, inner diameter 1.67 mm, outer diameter 2.42 mm, Clay-Adam, Parsippany, N.J., USA) with physiological saline (3 mg kg -1 ) containing Na 2 51 CrO 4 (0.5 Ci ml -1 ) and 10% charcoal.…”

“…CCKA receptors are found mainly in peripheral tissues and CCKB receptors express mainly in the central nervous system [21,24,42,47,56]. A number of studies have suggested that CCKA receptors play an important role in the inhibition of gastric emptying by amphetamine or endogenous and exogenous CCK [3,8,51]. However, the mechanism by which OT inhibits gastric activity through CCK is unknown.…”

“…The small intestine was then divided into ten equal segments, and the radioactivity in the stomach and each segment of small intestine was measured using an automatic gamma counter (1470 Wizard; Pharmacia). Gastric emptying was measured by determining the amount of labeled chromium contained in the small intestine 15 min after gavage expressed as a percentage of the amount given (Holzer 1985, Doong et al 1998.…”

“…Gastric emptying and gastrointestinal transit were measured as described by Doong et al (1998). The rats were fed by gavage via a catheter (PE-205, inner diameter (ID) 1.67 mm, outer diameter (OD) 2.42 mm, Clay-Adam, Parsippany, NJ, USA) with physiological saline (3 ml/kg) containing Na 2 51 CrO 4 (0.5 mCi/ml) and 10% charcoal, which was stirred continuously before use.…”
