INFUSIONS OF hFSH AND hLH IN NORMAL MEN

Abstract: Serial serum assays of immunoreactive FSH before, during and after a 4 h intravenous infusion of human follicle stimulating hormone (hFSH) in five healthy men revealed two disappearance rate constants with corresponding mean half-lives of 2.9 and 50.6 h. The mean distribution spaces calculated for the fast and slow component were 4.36 I and 75.9 1 respectively. The average value for the metabolic clearance rate was 17.2 ml/min and for the 225 Acta endocr. 81, 2 là I. Kinetics of human follicle stimulating horm… Show more

“…One protocol variation should be noted. We began LH infusions at 09.00 h while Kjeld et al (1976) started gonadotrophin administration between 12.00 and 14.00 h. We have preliminary data suggesting that in men Leydig cell responsiveness is different in the morning than it is in the afternoon.…”

“…This topic was reviewed by Mahoudeau et al (1975). Kjeld et al (1976) adminstered hLH alone to two men, and hLH with hFSH on four occasions begin¬ ning after 12.00 h. Twenty-five per cent of the gonadotrophin was given as a priming dose and the remainder was given as a 4 h infusion. During the infusion circulating LH rose more than 18 times basal as measured by radioimmunoassay.…”

“…Previous studies investigating FSH pharmacokinetics in the human male or female (Amin & Hunter, 1970;Kjeld et al, 1976;Diczfalusy & Harlin, 1988) had been complicated by the continuous secretion of endogenous FSH. Since immunoassays fail to distinguish between endogenous FSH and the injected human FSH preparation used in these studies, interference by the endogenous FSH, at least in the decay phase, is likely.…”
