LH is recognized as the Leydig cell stimulus for testosterone production. Yet earlier studies of bolus administered human pituitary LH did not result in testosterone elevations and spontaneous LH elevations infrequently cause recognizable parallel rises of testosterone in adult males. Therefore, 10 normal adult volunteers were given 3 h infusions of human pituitary LH A-3 (2 IU/kg) and sampling was continued for 3 h after the gonadotrophin was stopped. Testosterone, the precursor 17 OH-progesterone and LH were determined in blood samples. There was a mean 5.2-fold LH elevation. From a group baseline mean of 698 ng/100 ml \ m=+-\ 72 (\ m=+-\se) testosterone gradually rose and was significantly elevated at 90, 120 and 150 min (787 \m=+-\67 P <0.03, 767 \m=+-\60 P < 0.04, and 804 \ m=+-\69 P < 0.001, respectively). 17 OH\x=req-\ progesterone was determined on 9 subjects and averaged 142 ng/100 ml \m=+-\16 before LH and gradually rose to 178 \m=+-\18 at 120 min (P < 0.05) and then declined slightly at 150 and 180 min. Both steroids significantly declined between 180 min (end of LH infusion) and 240 min with no further significant concentration changes. The two men with the highest basal testosterone titers showed the smallest testosterone increases, but in both 17 OH-progesterone rose. In conclusion tonic LH stimulation caused mean testosterone and 17 OH-progesterone to peak at 150 min and at 120 min, respectively. Factors within the Leydig cell appear to limit increases of testosterone and 17 OH-progesterone during the first few hours of enchanced gonadotrophin stimulation.

SUMMARY The pharmacokinetics of a human FSH preparation widely used in clinical practice was evaluated in healthy male volunteers whose endogenous FSH was suppressed by 19‐nortestosterone injections. 9‐7 h following intramuscular injection of 150 IU FSH the mean immunoreactive peak concentration over baseline was 3–8 IU/1. Injection of 450 IU FSH led to a mean immunoreactive peak level of 10‐4 IU/1 at 9‐8 h. Mean half‐life was 24‐6 h (150 IU FSH) and 36‐2 h (450 IU FSH). Serum bioFSH concentrations were suppressed below the detection limit after injection of the 19‐nortestosterone and rose to preinjection levels after administration of FSH. Mean bioactive peak level for the 450 IU FSH was 7‐3 IU/1 FSH at 7‐4 h. The half‐life of bioFSH was calculated to be 13‐4 h and is considerably shorter than the half‐life of immunoFSH. Serum inhibin levels were suppressed by 19‐nortestosterone and did not change significantly after injection of the FSH preparation, suggesting that the FSH dose was insufficient to elicit this biological response. It is concluded that the currently used dose regimen for the treatment of male hypogonadism is inadequate for maintenace of serum bioFSH levels in the normal range.

ABSTRACT:The present study was designed to evaluate the protective effect of omega-3 against β-cyfluthrin (β-cyf)-induced neurotransmitters' impairment, oxidative damage, testicular toxicity and genotoxicity. Adult male Wistar rats were divided equally into four groups: Group (I): Control, Group (II): β-cyf (5 mg/kg body weight/day), Group (III): omega-3 (400 mg/kg body weight/da y) + β-cyf (5 mg/kg body weight/day), Group (IV): omega-3 (400 mg/kg body weight/day). β-cyf's treatment for 8 weeks inhibited the activities of AChE, DA, LDH, total ATPase and Na + /K + ATPase. Also, β-cyf resulted in an increase in oxidative stress as indicated by elevations in the levels of MDA and NO while, the levels of TAC, thiol content and TP were decreased in brain and testis. Sperm count, sperm motility and testosterone levels were reduced while, sperm abnormalities, FSH and LH levels were increased in β-cyf-treated group. Also, β-cyf administration led to a significant increase in TNFα, IL-6 and APP mRNA expressions. In addition, β-cyf caused histopathological alterations in brain and testis. The present results showed that omega-3 could improve all the measured parameters. In conclusion, omega-3 could exhibit protective effects against β-cyf induced neurological and testicular damage.
