Influence of Tangeretin on Tamoxifen's Therapeutic Benefit in Mammary Cancer

Abstract: We describe an in vivo model to study potential interference of dietary compounds, such as flavonoids, with tamoxifen, which could lead to reduced efficacy of adjuvant therapy. In our study, the tumor growth-inhibiting effect of oral tamoxifen was reversed upon addition of tangeretin to the diet. Our data argue against excessive consumption of tangeretin-added products and supplements by patients with mammary cancer during tamoxifen treatment.

“…The inverted microscope was connected with an MTI CCD72 camera (Dage MTI, Michigan City, IN, USA) and a U-matic VO-5850P videorecorder (Sony, Tokyo, Japan) at 30 s intervals. Track lengths were measured as described (Bracke et al, 1997). Haptotaxis assays (Aznavoorian et al, 1990) were performed in 48-well microchemotaxis chambers (NeuroProbe Inc., Bethesda, MD, USA) with 8-mm pore size Nucleopore filters (Costar, Cambridge, MA, USA).…”

“…It should, however, be recognized that the assay fails to encompass all the elements of the microecosystem present in natural tumors, environments where for example immunological factors can influence the invasive behavior of the cancer cells. In at least one study, the absence of such factors in the assay has led to conflicting results between the outcomes of the chick heart assay 29 and those of an animal model 30 .…”

“…The blocking antibody specific for human E-cadherin, mouse mAb SHE78-7 (Takara Shuzo Co., Kyoto, Japan), was used at 2 mg/ml to disrupt E-cadherin mediated cell-cell adhesion. Bracke et al (1997) described the possibility that MTT assay might be modified without alteration of growth. We examined cell growth, defined as the final number of DLD-1 cells remaining after 1-3 days in culture, both by directly counting the number of cells after trypsin detachment and by MTT assay.…”
